目的探讨低氧诱导因子-lα(HIF-1α)在视网膜母细胞瘤(Rb)中的表达及其与血管生成的关系。方法利用免疫组织化学方法检测60例Rb、10例正常视网膜组织中HIF-1α的表达,并用CD34单克隆抗体标记血管内皮细胞,计数Rb中微血管密度(MVD)。结果HIF-1α在正常视网膜组织中未见表达,主要表达于Rb细胞浆及胞核。HIF-lα在Rb组中阳性表达率为56.7%(34/60),明显高于正常对照组(P<0.01);Rb组中MVD平均为33.59±1.15,正常对照组为7.42±0.95(t=17.499,P<0.01)。HIF-lα与MVD的表达呈显著正相关(r=0.439,P<0.01)。结论HIF-lα在视网膜母细胞瘤中的表达与肿瘤的新生血管形成有关。提示HIF-lα的表达水平可用于预测Rb的生物学行为,可作为Rb治疗的新靶点。
Objective To investigate the expression of hypoxia inducible factor-1α (HIF-1α) in retinoblastoma (Rb) and its relationship with angiogenesis. Methods Immunohistochemistry was used to detect the expression of HIF-1α in 60 cases of Rb and 10 cases of normal retina. The CD34 monoclonal antibody was used to mark the vascular endothelial cells and the MVD was counted. Results HIF-1α was not expressed in normal retina and mainly expressed in the cytoplasm and nucleus of Rb. The positive expression rate of HIF-1α in Rb group was 56.7% (34/60), which was significantly higher than that in normal control group (P <0.01). The average MVD in Rb group was 33.59 ± 1.15 and that in normal control group was 7.42 ± 0.95 (t = 17.499, P <0.01). There was a significant positive correlation between the expression of HIF-1alpha and MVD (r = 0.439, P <0.01). Conclusion The expression of HIF-1α in retinoblastoma is related to neovascularization of tumor. It is suggested that the expression level of HIF-1αcan be used to predict the biological behavior of Rb, which can be used as a new target of Rb treatment.
