目的探讨β2肾上腺素受体基因(β2AR)Gln27Glu多态性对健康青年血脂及载脂蛋白比值的影响及在高糖低脂(HC/LF)膳食诱导的血脂及载脂蛋白比值变化中的作用。方法 56例健康志愿者,给予7d平衡膳食和6dHC/LF膳食,于第1d、第8d及第14d取12h空腹静脉血,测定血脂和载脂蛋白(apo)AI、B100水平,计算甘油三酯/高密度脂蛋白胆固醇(TG/HDL-C)、log(TG/HDL-C)、总胆固醇(TC)/HDL-C、低密度脂蛋白胆固醇(LDL-C)/HDL-C、apoAI/apoB100值。提取基因组DNA,聚合酶链反应-限制性酶切法分析β2ARGln27Glu位点多态性。结果血脂及载脂蛋白比值的基础值及平衡膳食后,无论是受试人群整体还是将男女分组分析,CC基因型受试者与G等位基因携带者之间均无明显差异。HC/LF膳食后,在整体受试人群中,G等位基因携带者log(TG/HDL-C)高于CC基因型受试者(P=0.038);男女分组分析发现CC基因型受试者与G等位基因携带者之间血脂及载脂蛋白比值无明显差异。与HC/LF膳食前相比,在受试人群整体,HC/LF膳食后apoAI/apoB100在CC基因型受试者升高(P=0.021),LDL-C/HDL-C和TC/HDL-C在CC基因型和G等位基因携带者中均下降(P<0.05)。按性别分组后,男性受试者HC/LF膳食后LDL-C/HDL-C和TC/HDL-C在CC基因型及G等位基因携带者均降低(P<0.05);女性CC基因型和G等位基因携带者HC/LF膳食后TG/HDL-C、log(TG/HDL-C)均升高(P<0.05),TC/HDL-C均降低(P<0.05),但LDL-C/HDL-C仅在CC基因型中降低(P=0.037)。结论β2AR基因Gln27Glu多态性G等位基因能够在健康青年女性抑制HC/LF膳食诱导的LDL-C/HDL-C降低。
Objective To investigate the effects of Gln27Glu polymorphism of β2AR on the ratio of serum lipids and apolipoproteins in healthy youth and the effect of high fat / low fat (HC / LF) diet on lipid and apolipoprotein ratio . Methods Fifty-six healthy volunteers were given 7d balanced diet and 6dHC / LF diet. Fasting venous blood was collected 12h and 12h after the first, eighth and fourteenth day respectively. Serum lipids and apolipoprotein AI and B100 levels were measured. Triglyceride (TG / HDL-C), total cholesterol (TC) / HDL-C, LDL-C / HDL-C and apoAI / apoB100 value. The genomic DNA was extracted and the polymorphism of β2ARGln27Glu site was analyzed by polymerase chain reaction - restriction enzyme digestion. Results The basic values of serum lipids and apolipoproteins, and the balanced diet, no significant difference between CC genotype subjects and G allele carriers, whether it is the overall population of the study population or the analysis of men and women. After HC / LF diet, the G allele carriers (log TG / HDL-C) were higher than CC genotype subjects in overall population (P = 0.038); CC genotype There was no significant difference in the ratio of blood lipids and apolipoproteins among carriers of G allele. Compared with the HC / LF diet, apoAI / apoB100 increased in subjects with CC genotype (P = 0.021), LDL-C / HDL-C and TC / HDL- C decreased in CC genotype and G allele carriers (P <0.05). LDL-C / HDL-C and TC / HDL-C were lower in CC genotypes and G allele carriers (P <0.05) after HC / LF diet in male subjects when grouped by sex; female CC genotype TG / HDL-C, log (TG / HDL-C) and TC / HDL-C were significantly lower in HC / LF and G allele carriers (P <0.05) -C / HDL-C decreased only in the CC genotype (P = 0.037). Conclusion G allele of Gln27Glu polymorphism of β2AR gene can inhibit the decrease of LDL-C / HDL-C induced by HC / LF diet in healthy young women.
