采用反相高效液相色谱(RP-HPLC)的方法对狭叶松果菊组培苗不同部位、愈伤组织以及不同株系毛状根中的松果菊苷和绿原酸进行了含量测定,建立同时测定狭叶松果菊组织培养物中松果菊苷和绿原酸含量的高效液相色谱方法;实验采用Kromasil ODS C18柱,流动相为乙腈∶甲醇∶0.1%磷酸溶液(10∶15∶75);检测波长为330nm。结果表明松果菊苷和绿原酸在0~25μg/mL范围线性关系良好;平均回收率方面松果菊苷为97.6%(RSD=0.83%),绿原酸为98.5%(RSD=0.91%)。该法简便、准确,可作为狭叶松果菊组织培养物中松果菊苷和绿原酸的定量分析方法。
The contents of echinacoside and chlorogenic acid in different parts of the plantlets of Echinacea purpurea, callus, and hairy roots of different strains were determined by reverse-phase high performance liquid chromatography (RP-HPLC) , A HPLC method for simultaneous determination of echinacosin and chlorogenic acid in the tissue culture of Echinacea purpurea was established. The experiment was performed on a Kromasil ODS C18 column with a mobile phase of acetonitrile: methanol: 0.1% phosphoric acid solution (10:15 : 75); detection wavelength is 330nm. The results showed that there was a good linear relationship between echinacoside and chlorogenic acid in the range of 0-25 μg / mL. The average recoveries of echinacoside were 97.6% (RSD = 0.83%), chlorogenic acid was 98.5% (RSD = 0.91% ). The method is simple and accurate and can be used as a quantitative method for the determination of echinacoside and chlorogenic acid in the tissue culture of Echinacea purpurea.
