目的:建立巴戟天及不同盐分炮制品中糖类成分HPLC-ELSD指纹图谱分析方法,为确定盐制巴戟天饮片在炮制过程中的用盐比例及其质量标准的制定提供依据。方法:采用HPLC,色谱柱为Agilent ZORBAX Carbohyrate(4.6 mm×250mm,5μm),流动相为乙腈-甲醇-水,流速1.2 mL.min-1,梯度洗脱,柱温35℃,以蒸发光散射检测器(ELSD)检测,漂移管温度为105℃,增益值为1,氮气流量2.0 L.min-1;采用国家药典委员会推荐的《中药色谱指纹图谱相似度评价系统》(2004年A版)进行评价。结果:以耐斯糖峰为参照峰,18批样品标定了共有峰16个,各样品相似度均>0.90。结论:该方法简单、准确、重复性好,可用于巴戟天及其盐制品糖类成分的质量控制。
OBJECTIVE: To establish HPLC-ELSD fingerprinting method for the determination of sugars in the processed products of Morinda officinalis and its salt, and provide the basis for the determination of salt proportion and its quality standard during the processing of Salvia miltiorrhiza. Methods: The HPLC was carried out on an Agilent ZORBAX Carbohyrate column (4.6 mm × 250 mm, 5 μm) with a mobile phase of acetonitrile-methanol-water at a flow rate of 1.2 mL · min-1 and gradient elution at 35 ℃. (ELSD), the temperature of drift tube was 105 ℃, the gain value was 1, the nitrogen flow rate was 2.0 L.min-1. According to the “Evaluation System of Similarity of Chromatographic Fingerprints of Traditional Chinese Medicine” (2004 Edition) recommended by the State Pharmacopoeia Commission, Evaluation. Results: With the peak of Nesugose as the reference peak, 16 batches of samples were calibrated in 18 batches of samples, and the similarity of each sample was> 0.90. Conclusion: The method is simple, accurate and reproducible. It can be used for the quality control of sugars in Morinda officinalis and its salt products.
