目的:研究大孔树脂分离纯化金荞麦抗癌成分的工艺。方法:以原花青素B2吸附率和解析率为指标,对七种树脂进行静态筛选,采用正交试验法,考察最佳上样条件,系统研究洗脱参数。结果:X-5型树脂对原花青素B2有较好的吸附分离性能,最佳吸附条件:3.5BV浓度为0.12g·mL-1的生药,pH为3.0,流速为2BV·h-1;最佳解吸条件:解吸液为3BV60%乙醇水溶液,流速为2 BV·h-1。结论:该工艺用于金荞麦抗癌成分的纯化是可行的。 Objective: To study the technology of separating and purifying anticancer components of Fagopyrum tataricum with macroporous resin. Methods: Seven kinds of resins were screened by the adsorption rate and resolution of proanthocyanidins B2. The orthogonal test was used to investigate the optimal loading conditions and the elution parameters were systematically studied. Results: X-5 resin had good adsorption and separation performance for proanthocyanidin B2. The optimal adsorption conditions were as follows: the crude drug with 3.5BV concentration was 0.12g · mL-1, the pH was 3.0 and the flow rate was 2BV · h-1; Desorption conditions: desorption solution 3BV60% ethanol solution, the flow rate of 2 BV · h-1. Conclusion: The process is feasible for the purification of anti-cancer components of buckwheat.
