目的建立高效毛细管电泳二极管阵列检测法(HPCE-DAD)同时测定不同产地、采收期和种属的罗布麻叶中芦丁、异槲皮苷、金丝桃苷及槲皮素含量的方法。方法选择毛细管区带电泳分离模式,以45mmol.L-1硼砂缓冲液(pH9.2)为电泳介质,未涂渍标准熔融石英毛细管(64.5cm×75μm,有效长度56cm)为分离通道,分离电压为25kV,检测波长为206nm,毛细管温度为25℃,压力进样为5000Pa,5s。结果4种黄酮的峰面积与浓度的线性关系良好(r>0.9986);加样回收率为95.34%~102.65%。结论该方法简单、准确,重现性较好,可用于罗布麻叶药材内在质量的评价和控制。
OBJECTIVE To establish a method for the simultaneous determination of rutin, isoquercitrin, hyperoside and quercetin in Apocynum leaves collected from different habitats, harvest periods and species by high performance capillary electrophoresis with diode array detection (HPCE-DAD). Methods The capillary zone electrophoresis separation mode was selected. The standard fused silica capillary (64.5cm × 75μm, effective length 56cm) was used as the separation channel with 45mmol.L-1 borax buffer (pH 9.2) as the electrophoresis medium. The separation voltage For 25kV, detection wavelength of 206nm, capillary temperature of 25 ℃, pressure injection of 5000Pa, 5s. Results The linear relationship between the peak area and the concentration of the four flavonoids was good (r> 0.9986). The recoveries of the four flavonoids were 95.34% -102.65%. Conclusion The method is simple, accurate and reproducible. It can be used to evaluate and control the intrinsic quality of Apocynum venetum leaves.
