目的分析螨变应原的聚类、所属的蛋白家族及其氨基酸序列组成特点。方法从国际免疫学会联合会变应原命名数据库获得螨类变应原氨基酸序列,采用生物信息学方法进行进化树构建、蛋白质家族和超家族分类、序列比对、二级结构分析和序列相似度分析。结果螨类变应原第1~24组分在系统进化树中按照功能聚类成7大簇,归属于Trypsin等16个蛋白质家族和E set domains等13个蛋白质超家族。通过序列比对,获得螨类变应原第1、2组分的一致性氨基酸和独有氨基酸信息及第1、2组分亚型间氨基酸置换信息。螨类变应原第1、2组分的二级结构基本都由α-螺旋、延伸主链和无规卷曲构成,但Tyr p 2的二级结构不含α-螺旋。Der f 1、Der p1和Eur m 1序列相似度高(82.77%~84.50%),在进化树中也聚类在一起;Der f 2、Der p 2和Eur m 2亦然(相似度分别为84.17%~85.15%)。结论螨类变应原第1~24组分归属于16个蛋白质家族和13个蛋白质超家族,并聚类为7个簇,每簇变应原都有其特定功能,可为新变应原的寻找和变态反应学的基础研究提供参考,并为重组变应原的研究提供新的方向。
Objective To analyze the clustering of mite allergens, their protein families and their amino acid sequences. Methods The amino acid sequence of mite allergen was obtained from the International Federation of Immunology Societies allergen database. The bioinformatics method was used to construct phylogenetic tree, classification of protein family and superfamily, sequence alignment, secondary structure analysis and sequence similarity analysis. Results The mite allergen components 1 ~ 24 were clustered into 7 clusters in the phylogenetic tree, belonging to 16 protein families such as Trypsin and 13 protein superfamilies such as E set domains. Through sequence alignment, we got the consistent amino acid and unique amino acid information of the first and the second component of mite allergens and the amino acid substitution information between the first and the second component subtypes. The secondary structure of mite allergen components 1 and 2 are basically composed of α-helix, extending backbone and random coil, but the secondary structure of Tyr p 2 does not contain α-helix. Der f 1, Der p1 and Eur m 1 sequences were highly homologous (82.77% -84.50%) and clustered together in the phylogenetic tree; Der f 2, Der p 2 and Eur m 2 were also similar 84.17% ~ 85.15%). CONCLUSIONS The first to 24th components of the mite allergen belong to 16 protein families and 13 protein superfamilies and are clustered into 7 clusters. Each allergen has its own specific function and may be a new allergen Search and find the basic research of allergy to provide a reference, and provide a new direction for the study of recombinant allergens.
