目的 :探讨Rho相关卷曲螺旋形成蛋白激酶1(ROCK1)在肝癌中的表达及在肝癌侵袭中的作用。方法 :荧光定量PCR(real-time PCR)检测肝癌及癌旁组织中ROCK1的mRNA表达水平,同时检测肝癌细胞系及正常人肝细胞中ROCK1mRNA表达水平,免疫组织化学染色(IHC)法检测肝癌及癌旁组织中ROCK1蛋白的表达量,并通过在肝癌细胞株中过表达及干扰ROCK1分析ROCK1蛋白对肝癌细胞转移的作用。结果:60对样本中有42对ROCK1的mRNA表达水平在肝癌组织中高于癌旁组织,免疫组织化学染色提示肝癌组织中ROCK1蛋白表达量高于癌旁组织。在体外肝癌细胞的功能实验中,过表达ROCK1能促进HepG2细胞的侵袭和转移,而干扰ROCK1的表达能减少SMMC-7721细胞的侵袭和转移。结论:ROCK1在肝癌组织中高表达,过表达ROCK1能促进肿瘤的侵袭及转移,ROCK1可能成为临床治疗中抑制肿瘤侵袭和转移的靶点。
Objective: To investigate the expression of Rho-associated coiled-coil-forming protein kinase 1 (ROCK1) in hepatocellular carcinoma and its role in the invasion of hepatocellular carcinoma. Methods: The mRNA expression of ROCK1 in hepatocellular carcinoma and its adjacent tissues was detected by real-time PCR. The expression of ROCK1 mRNA in hepatocellular carcinoma cell lines and normal human hepatocytes was detected simultaneously. The expression of ROCK1 mRNA was detected by IHC The expression of ROCK1 protein in paracancerous tissues and the effect of ROCK1 protein on the metastasis of hepatoma cells were analyzed by Overexpression of ROCK1 and interference with ROCK1 in hepatocellular carcinoma cell lines. Results: The mRNA expression of ROCK1 in 42 pairs of samples was higher than that in paracancerous tissues in 60 pairs of samples. Immunohistochemical staining indicated that the expression of ROCK1 protein in HCC tissues was higher than that in paracancerous tissues. Overexpression of ROCK1 can promote the invasion and metastasis of HepG2 cells in vitro. However, the interference of ROCK1 expression can reduce the invasion and metastasis of SMMC-7721 cells. CONCLUSION: ROCK1 is overexpressed in HCC tissues. Overexpression of ROCK1 can promote tumor invasion and metastasis, and ROCK1 may be the target of inhibiting tumor invasion and metastasis in clinical treatment.
