目的探讨兔肝VX2移植瘤治疗前后磁共振背景信号抑制弥散加权体部成像(diffusion weighted imagingwith background suppression,DWIBS)特征,对照分析肿瘤表观扩散系数(apparent diffusion coefficient,ADC)值与肿瘤细胞密度是否存在相关性。方法兔肝VX2移植瘤模型随机分为三组(8只/组)。建模后12天:TACE+ES组行TA-CE治疗和内皮抑素治疗;内皮抑素组注射内皮抑素治疗12天;对照组注射生理盐水12天。所有动物模型均于治疗前1天、治疗后第13天行DWIBS检查并记录各组肿瘤实质的ADC值。MR检查后取出肿瘤组织行HE染色并计算细胞密度。结果治疗后对照组ADC值低于TACE+ES组和内皮抑素组并有统计学意义(P=0.008,P=0.001);ADC值与肿瘤细胞密度间存在负相关性(r=-0.844,P=0.000)。结论 DWIBS结合ADC值的定量测量可以对活体肿瘤细胞密度进行无创性动态评价,有望为在体肿瘤分级提供新的帮助。
Objective To investigate the characteristics of diffusion weighted imaging with background suppression (DWIBS) before and after treatment of rabbit VX2 tumor in vitro and to compare whether the apparent diffusion coefficient (ADC) value of tumor and tumor cell density There is a correlation. Methods Rabbit liver VX2 xenograft models were randomly divided into three groups (n = 8). At 12 days after modeling, TA-CE and endostatin were given to TACE + ES group. Endostatin was given to endostatin for 12 days and saline was injected into control group for 12 days. All animal models were on the 1st day before treatment and on the 13th day after treatment, DWIBS were performed and the ADC value of the tumor in each group was recorded. Tumor tissue was removed after MR examination and HE staining was performed and cell density was calculated. Results The ADC value of control group was lower than that of TACE + ES group and endostatin group after treatment (P = 0.008, P = 0.001). There was a negative correlation between ADC value and tumor cell density (r = -0.844, P = 0.000). Conclusion The quantitative measurement of DWIBS combined with ADC value can evaluate noninvasive dynamic cell density of living tumor cells and may provide new help for the classification of tumor in vivo.
