目的 建立厌氧罐法检查硫乙醇酸盐流体培养基灵敏度的方法。方法 用氧化还原指示剂检查厌氧罐的密封性能 ,确保厌氧罐内能达到厌氧状态 ;测定达到厌氧状态的时间以及能够保持的时间 ;将生孢梭菌接种至不同的固体培养基上 ,根据菌落生长情况选择适宜生孢梭菌生长的培养基 ;同时用厌氧罐法和试管法对同一批号的生孢梭菌菌悬液培养计数 ,并对计数结果进行比对。结果 硫乙醇酸盐流体培养基能够作为厌氧罐内厌氧状态的指示剂 ,罐盖密封后 3 0min可达厌氧状态 ,至少可以保持7天 ;中国药典规定的营养琼脂培养基适宜生孢梭菌的生长 ;厌氧罐法比试管法更适合对生孢梭菌菌悬液的培养计数。结论 用厌氧罐法检查硫乙醇酸盐流体培养基的灵敏度能更真实地反映培养基的质量 ,符合中国药典要求 ,方法切实可行
Objective To establish an anaerobic tank method to check the sensitivity of thioglycollate fluid medium. Methods The redox indicator was used to check the sealing performance of the anaerobic tank to ensure that the anaerobic tank could reach the anaerobic status. The time to reach the anaerobic status and the time it could be maintained were determined. Clostridium sporogenes were inoculated into different solid medium On the basis of the growth of colony selection for Clostridium sporotrichum growth medium; the same batch with anaerobic tank method and test tube number of the same batch of Clostridium sp. Clostridium suspension culture count, and the count results were compared. Results Thioglycollate fluid medium can be used as an anaerobic tank anaerobic status indicator, the lid sealed 30min anaerobic state, at least for 7 days; Chinese Pharmacopoeia nutrient agar culture suitable for spores Clostridium growth; anaerobic tank test tube method is more suitable for culture of Clostridium sp. Clostridium suspension culture count. Conclusion The anaerobic tank method to check the sensitivity of thioglycolic fluid medium can more accurately reflect the quality of the medium, in line with Chinese Pharmacopoeia requirements, the method is feasible
