目的:采用切除双侧卵巢复制骨质疏松症大鼠模型,观察补肾复方对去卵巢大鼠骨质疏松症骨生物力学、骨微结构、骨钙素(BGP)和抗酒石酸盐酸性磷酸酶(TRACP)的影响。方法:将36只SD雌性大鼠随机分为假手术组、模型组和中药组3组。切除双侧卵巢复制骨质疏松症大鼠模型,造模成功后,中药组给予相应药物,灌胃3个月后取材。三点弯曲实验获得骨生物力学相关数据;Micro CT测量获得骨矿物密度和骨小梁结构相关参数;ELISA试剂盒检测血清BGP及TRACP含量。结果:与假手术组比较,模型组最大应力、弹断裂吸收能量均明显下降,差异具有统计学意义(P<0.05);模型组的骨矿物密度(BMD)、骨体积/总体积(BV/TV),骨小梁厚度(Tb.Th),骨小梁数目(Tb.N)均明显降低,差异具有统计学意义(P<0.05);骨小梁分离度(Tb.Sp)明显升高,差异具有统计学意义(P<0.05);模型组的血清BGP及TRACP均明显升高,差异具有统计学意义(P<0.05)。与模型组比较,中药组最大应力和断裂吸收能量均明显升高,差异具有统计学意义(P<0.05);中药组的BMD、BV/TV、Tb.Th、Tb.N无明显差异,但均有增长的趋势,Tb.Sp有降低的趋势;中药组的血清BGP及TRACP含量均明显降低,差异具有统计学意义(P<0.05)。结论:补肾复方可能通过降低OP模型大鼠血清TRACP、BGP含量,提高骨生物力学性能和改善骨结构退行性改变,达到治疗骨质疏松症的作用。
OBJECTIVE: To observe the effects of Bushen Compound on the osteoporosis osteoblastic biomechanics, bone microarchitecture, BGP and tartrate-resistant acid phosphatase in ovariectomized rats with osteoporosis TRACP) effect. Methods: Thirty-six SD female rats were randomly divided into sham operation group, model group and TCM group. After ovariectomized osteoporosis rat model was excised, the Chinese medicine group was given the appropriate medicine after successful modeling. The rats were drawn for 3 months after gavage. Three-point bending test was used to obtain the biomechanical data. The bone mineral density and trabecular structure parameters were obtained by Micro CT. The contents of serum BGP and TRACP were detected by ELISA kit. Results: Compared with the sham operation group, the maximal stress and the energy absorbed by the bullet fracture in the model group decreased significantly (P <0.05). The bone mineral density (BMD), bone volume / total volume (BV / TV, trabecular thickness, Tb.N were significantly decreased (P <0.05), and Tb.Sp was significantly increased , The difference was statistically significant (P <0.05). The serum BGP and TRACP in the model group were significantly increased, the difference was statistically significant (P <0.05). Compared with the model group, the maximum stress and the energy absorbed by rupture in the Chinese herbal medicine group were significantly increased (P <0.05), while there was no significant difference in BMD, BV / TV, Tb.Th and Tb.N in the Chinese herbal medicine group (P <0.05). The levels of BGP and TRACP in the TCM group were significantly lower than those in the control group (P <0.05). Conclusion: Bushen Compound may reduce the serum TRACP, BGP content, improve the biomechanical properties of bone and improve the degenerative changes of bone structure in rats with OP, so as to treat osteoporosis.
