目的 探讨实验性铅中毒对睾丸组织神经生长因子 (NGF)基因表达的影响。 方法 小鼠以 0 .5 %醋酸铅自由饮服 8周 ,测定血铅和睾丸铅含量。采用原位杂交法 (以地高辛标记 NGF DNA为探针 )和 RT- PCR方法 ,观察睾丸组织 NGF m RNA含量的变化。 结果 铅染毒小鼠血铅和睾丸组织铅含量明显升高 ;原位杂交结果显示 ,铅染毒小鼠睾丸杂交信号明显减少 ;RT- PCR结果表明 ,铅染毒小鼠睾丸组织 NGF m RNA表达量明显下降。 结论 铅中毒可使睾丸组织 NGF基因表达量减少
Objective To investigate the effects of experimental lead poisoning on the expression of nerve growth factor (NGF) gene in testis. Methods The mice were treated with 0.5% lead acetate for 8 weeks, and the contents of lead in blood and testis were determined. In situ hybridization (digoxigenin-labeled NGF DNA probe) and RT-PCR method were used to observe the changes of testicular tissue NGF m RNA. Results Lead levels in blood lead and testis tissue were significantly increased in lead-poisoned mice. In situ hybridization results showed that testis hybridization signals in lead-exposed mice were significantly reduced. RT-PCR results showed that lead-poisoned mice testicle tissue NGF m RNA The amount of expression decreased significantly. Conclusion Lead poisoning can reduce the expression of NGF gene in testis tissue
