目的 观察转化生长因子β(transforming grow th factor-beta,TGF-β)和干扰素γ(interferon-gam m a,IFN-γ)对人视网膜色素上皮细胞(retinalpigm entepithelialcells,RPE)胶原合成的影响。 方法 用3H-脯氨酸掺入、免疫细胞化学及分子杂交技术观察TGF-β和IFN-γ分别及联合作用下培养的人RPE胶原合成活性。 结果 TGF-β在10 ng/ml浓度使RPE对3H-脯氨酸的摄取增加130.87% ,并增强Ⅳ、Ⅰ和Ⅲ型胶原荧光染色和m RNA表达。IFN-γ在10 000 U/m l浓度抑制54.72% 的脯氨酸摄取,并减弱Ⅳ型胶原染色和3种胶原的m RNA表达。 结论 TGF-β和IFN-γ对RPE的胶原合成分别具有正向及负向调控作用,IFN-γ可能用于抑制RPE胶原合成。
Objective To observe the effects of transforming growth factor-β (TGF-β) and interferon-γ (IFN-γ) on the collagen synthesis of human retinal pigment epithelial cells (RPE). Methods 3H-Proline incorporation, immunocytochemistry and molecular hybridization were used to observe the collagen synthesis activity of human RPE cultured in vitro and in combination with TGF-β and IFN-γ respectively. Results TGF-β increased the uptake of 3H-proline by 130.87% in 10 ng / ml RPE, and enhanced the expression of collagen type IV, Ⅰ and Ⅲ and m RNA. IFN-γ inhibited proline uptake by 54.72% at a concentration of 10,000 U / ml and attenuated type IV collagen staining and mRNA expression of three types of collagen. Conclusion TGF-β and IFN-γ have positive and negative regulatory effects on collagen synthesis of RPE, IFN-γ may be used to inhibit RPE collagen synthesis.
