目的优选从甘草不同器官中提取高质量RNA的方法,为研究甘草药用成分合成酶基因克隆和基因表达奠定基础。方法比较异硫氰酸胍法、柠檬酸钠法所提取的甘草根木质部、根皮部、根茎、叶中RNA的纯度和得率。结果异硫氰酸胍法能从根的木质部、根皮部和根茎中提取出质量较好的RNA,但不能从甘草叶片中提出RNA;柠檬酸钠法能从叶片中提取出质量较高的RNA。结论异硫氰酸胍法和柠檬酸钠法分别是提取甘草根木质部、根皮部和根茎及叶片中RNA的有效方法。
Objective To optimize the extraction of high quality RNA from different organs of Glycyrrhiza uralensis, and to lay a foundation for the study of cloning and gene expression of component synthetase genes in Glycyrrhizae. Methods The purity and yield of RNA in xylem root, root bark, rhizome and leaf of licorice root were extracted by guanidinium thiocyanate method and sodium citrate method. Results The guanidinium thiocyanate method can extract RNA with good quality from the xylem root, root bark and rhizome of roots, but it cannot extract RNA from licorice leaves; sodium citrate method can extract higher quality from leaves. RNA. Conclusion The guanidinium isothiocyanate method and the sodium citrate method are effective methods for the extraction of RNA in xylem root, root bark, rhizome and leaf, respectively.
