目的探讨平喘益肺合剂对哮喘大鼠气道重塑和α-平滑肌肌动蛋白(α-SMA)的影响及可能的作用机制。方法选取40只雄性SD大鼠,随机分为正常对照组(A组)、哮喘组(B组)、地塞米松组(C组)、平喘益肺合剂组(D组),每组各10只。用卵白蛋白致敏吸入激发制备哮喘模型。在哮喘激发4周后测量肺组织α-SMA mRNA和气道壁Ⅲ型胶原蛋白的表达。结果 B组α-SMA mRNA、Ⅲ型胶原表达明显高于A组(P<0.05);C组和D组α-SMA mRNA、Ⅲ型胶原表达显著低于B组(P<0.05)。结论平喘益肺合剂通过降低α-SMA mRNA的过度表达,抑制Ⅲ型胶原的沉积,减轻气道重塑的发生。
Objective To investigate the effect of Pingchuan Yifei Mixture on airway remodeling and α-SMA in asthmatic rats and its possible mechanism. Methods Forty male Sprague Dawley rats were randomly divided into four groups: normal control group (group A), asthma group (group B), dexamethasone group (group C) and Pingchuan yifei mixture group (group D) 10 only Asthma models were prepared by inhalation challenge with ovalbumin. The expression of α-SMA mRNA and type III collagen in airway wall was measured 4 weeks after asthma challenge. Results The expression of α-SMA mRNA and type Ⅲ collagen in group B was significantly higher than that in group A (P <0.05). The expressions of α-SMA mRNA and type Ⅲ collagen in group C and group D were significantly lower than those in group B (P <0.05). Conclusion Pingdu Yifei Mixture can reduce the deposition of type Ⅲ collagen and reduce the occurrence of airway remodeling by decreasing α-SMA mRNA overexpression.
