目的探讨半枝莲提取物(SBE)对LoVo细胞增殖及侵袭能力的影响及其可能机制。方法分别用1.1,2.2mg/ml的SBE处理LoVo细胞,以不加SBE的LoVo细胞作对照,同时以5-氟尿嘧啶(5-FU)5μg/ml作为阳性对照组(5-FU组)。MTT法检测处理后72h内细胞增殖能力的变化,细胞克隆形成实验和侵袭实验分别观察细胞克隆形成能力和侵袭能力的变化,Westernblot检测细胞Twist和E-钙粘素蛋白的表达。结果与对照组相比,5-FU组和SBE高剂量组的LoVo细胞增殖能力、克隆形成能力和侵袭能力均明显降低,E-钙粘素表达增加,Twist蛋白表达减少(P<0.05)。结论2.2mg/ml的SBE对LoVo细胞增殖和侵袭能力的抑制作用可能与上调E-钙粘素、下调Twist蛋白的表达有关。
Objective To investigate the effects of Scutellaria barbata extract (SBE) on proliferation and invasion of LoVo cells and its possible mechanism. Methods LoVo cells were treated with SBE at 1.1 and 2.2 mg / ml, respectively. LoVo cells without SBE were used as controls, and 5-FU (5-FU) 5 μg / ml was used as positive control. The proliferation of cells was detected by MTT assay within 72h. The changes of cell colony-forming ability and invasion ability were observed by cell clone formation assay and invasion assay. The expressions of Twist and E-cadherin were detected by Western blot. Results Compared with the control group, the proliferation, clonogenicity and invasiveness of LoVo cells in 5-FU and SBE high-dose groups were significantly decreased, while the expression of E-cadherin and Twist decreased (P <0.05). Conclusion The inhibitory effect of 2.2mg / ml SBE on the proliferation and invasion of LoVo cells may be related to up-regulation of E-cadherin and down-regulation of Twist protein.
