目的 探讨榄香烯对小鼠肝癌腹水瘤细胞系Hca F2 5/CL 1 6A3治疗的作用机制。方法 将 2 2只小鼠分成两组 ,对照组 1 2只 ,治疗组 1 0只。从荷瘤次日起 ,治疗组用榄香烯腹腔注射治疗 ,对照组用生理盐水治疗。 2 0d后取瘤组织用免疫组织化学染色方法观察荷瘤小鼠两组肿瘤组织的视网膜母细胞瘤抑癌蛋白 (pRB)和腺病毒E2启动子结合因子 (E2F 1 )蛋白的表达情况。结果 对照组肿瘤组织 pRB阳性率为 50 % (6/ 1 2 ) ,用药组阳性率为 1 0 0 % (1 0 / 1 0 ) ,两组间差异有显著性 (P <0 .0 5)。对照组和用药组E2F 1均呈阳性表达 ,表达率均为 1 0 0 % (1 2 / 1 2 )和 (1 0 / 1 0 ) ,两组间差异无显著性 (P >0 .0 5)。结论 榄香烯对小鼠肝癌腹水瘤细胞系Hca F2 5/CL 1 6A3有明显的抑制作用。其作用机制与 pRB蛋白表达有关 ,与E2F 1无关
Objective To investigate the mechanism of elemene treatment of mouse hepatocellular carcinoma cell line Hca F2 5 / CL 1 6A3. Methods Twenty-two mice were divided into two groups, 12 in the control group and 10 in the treatment group. From the next day after the tumor-bearing, the treatment group was treated with elemene intraperitoneally and the control group was treated with saline. Immunohistochemical staining was used to observe the expression of retinoblastoma suppressor protein (pRB) and adenovirus E2 promoter (E2F 1) protein in tumor tissues of the two tumor-bearing mice on day 20 after tumor formation. Results The positive rate of pRB in the control group was 50% (6/12) and the positive rate was 100% (10/10) in the control group, with significant difference between the two groups (P <0.05) . E2F 1 in the control group and the treatment group were all positive, the expression rates were 100% (1 2/1 2) and (1 0/1 0) respectively, with no significant difference between the two groups (P> 0.05) ). Conclusion Elemene significantly inhibits mouse hepatocellular carcinoma cell line Hca F2 5 / CL 1 6A3. Its mechanism of action is related to pRB protein expression, but not to E2F 1
