目的:观察C57BL/6小鼠肝脏中NK细胞、T细胞和NKT细胞的含量、亚型和细胞因子产生能力方法:分离正常C57BL/6小鼠肝脏的淋巴细胞,通过细胞表面分子染色以及流式细胞术(FCM)检测NK1.1~+NK细胞、CD3~+T细胞和CD3`+NK1.1~+NKT细胞的含量及其表面CD4、CD8分子的表达情况;细胞经过PMA+Ionomycin刺激后,应用细胞内细胞因子染色并结合FCM检测NK1.1~+NK细胞、CD3~+T细胞和CD3~+NK1.1~+NKT细胞产生IFN-γ、IL-4、IL-9和IL-17细胞因子的情况。结果:肝脏中NKT细胞的含量为(21.51±13.02)%,高于NK细胞和T细胞(P<0.05)。NK细胞以CD4~-CD8~-亚群为主,T细胞和NKT细胞均以CD4~+细胞亚群为主。NKT细胞分泌的IL-4的比例高于NK细胞和T细胞(P<0.05)。T细胞分泌IL-17的能力明显强于NK细胞和NKT细胞(P<0.05),IL-9~+细胞在三种细胞中含量不高(<0.03%)。结论:C57BL/6小鼠肝脏中NK细胞、T细胞和NKT细胞含量、表型和功能方面可能存在明显的差异
Objective: To observe the contents, subtypes and cytokines production of NK cells, T cells and NKT cells in the liver of C57BL / 6 mice. METHODS: Lymphocytes from the liver of normal C57BL / 6 mice were isolated and stained by cell surface molecules and flow cytometry The levels of NK1.1 ~ + NK cells, CD3 ~ + T cells and CD3 ’+ NK1.1 ~ + NKT cells and the expression of CD4 and CD8 on the surface of NK1.1 ~ + NK cells were detected by FCM. After stimulated by PMA + Ionomycin , IL-4, IL-9 and IL-9 were detected by intracellular cytokine staining combined with FCM in NK1.1 ~ + NK cells, CD3 ~ + T cells and CD3 ~ + NK1.1 ~ 17 cytokines. Results: The content of NKT cells in liver was (21.51 ± 13.02)%, higher than those in NK cells and T cells (P <0.05). The majority of NK cells were CD4 ~ -CD8 ~ - subpopulations, while T cells and NKT cells were predominantly CD4 ~ + subpopulations. NKT cells secreted higher proportion of IL-4 than NK cells and T cells (P <0.05). The ability of T cells to secrete IL-17 was significantly stronger than that of NK cells and NKT cells (P <0.05). The content of IL-9 ~ + cells in the three cells was not high (<0.03%). Conclusion: The content, phenotype and function of NK cells, T cells and NKT cells in liver of C57BL / 6 mice may be significantly different
