目的:探讨逆向诱导半胱氨酸丰富蛋白(RECK)与膜型基质金属蛋白酶-1(MT1-MMP)在膀胱尿路上皮癌(BTCC)发生发展中的作用。方法:采用标记试剂-量子点免疫荧光组织化学技术检测人BTCC组织芯片150芯(包括70例BTCC组织和5例膀胱炎组织)中RECK与MT1-MMP的表达,并分析RECK和MT1-MMP的表达与BTCC临床和病理特征的关系。结果:1)RECK在膀胱炎中呈高表达,平均光密度为0.457 1±0.031 2,阳性面积率为0.421 9±0.030 4;BTCC中RECK呈低表达,平均光密度为0.125 4±0.018 7,阳性面积率为0.125 3±0.017 4。RECK在膀胱炎中的表达显著高于BTCC,P<0.05。2)MT1-MMP在BTCC中呈高表达,MT1-MMP的平均光密度为0.452 0±0.031 6,阳性面积率为0.498 2±0.032 6;MT1-MMP在膀胱炎中呈低表达,平均光密度为0.165 1±0.017 9,阳性面积率为0.148 2±0.013 3。MT1-MMP在BTCC中的表达显著高于膀胱炎,P<0.05。3)RECK和MT1-MMP的表达与患者年龄、性别之间的差异均无统计学意义(P>0.05),但与肿瘤浸润深度、分化程度和临床UICC分期相关,差异均有统计学意义,P<0.05。结论:RECK在BTCC中呈低表达,MT1-MMP在BTCC中呈高表达,两者可能参与了BTCC的发生,并在其的侵袭和转移中发挥作用。
Objective: To investigate the role of reversely induced RECK and MT1-MMP in the development of bladder urothelial carcinoma (BTCC). METHODS: The expression of RECK and MT1-MMP in 150 core of human BTCC tissue (including 70 BTCC tissues and 5 cystitis tissues) was detected by using labeled reagent-QDIS. The expressions of RECK and MT1-MMP Relationship between expression and clinical and pathological features of BTCC. RECK was highly expressed in cystitis, the average optical density was 0.457 1 ± 0.031 2, the positive area rate was 0.421 9 ± 0.030 4; RECK was low expressed in BTCC with the average optical density of 0.125 4 ± 0.018 7, The positive area rate was 0.125 3 ± 0.017 4. The expression of RECK in cystitis was significantly higher than that in BTCC, P <0.05.2) MT1-MMP was highly expressed in BTCC, the average optical density of MT1-MMP was 0.452 ± 0.031 6, the positive area rate was 0.498 2 ± 0.032 6; MT1-MMP was low expression in cystitis, the average optical density was 0.165 1 ± 0.017 9, the positive area rate was 0.148 2 ± 0.013 3. The expression of MT1-MMP in BTCC was significantly higher than that in cystitis (P <0.05). There was no significant difference between the expression of RECK and MT1-MMP and the age and gender of patients (P> 0.05) The depth of invasion, the degree of differentiation and the clinical stage of UICC were significantly different (P <0.05). CONCLUSION: RECK is low expressed in BTCC and MT1-MMP is highly expressed in BTCC, both of which may be involved in the occurrence of BTCC and play a role in its invasion and metastasis.
