目的探讨利用聚合酶链反应(PCR)联合直接基因测序技术检测感染性眼内炎的可行性及有效性,为感染性眼内炎患者的快速诊断寻求一种新的检测方式。方法对我院2005年10月至2006年10月手术治疗的感染性眼内炎患者47例47眼,手术取材进行PCR联合直接基因测序检查并与细菌涂片、细菌培养、真菌培养、厌氧菌培养检查结果对比。结果PCR联合基因诊断方法检查时间较传统方法耗时少。PCR病原菌检测阳性率高,为25例,25例阳性者中经直接基因测序出病原体类型19例。与病原体培养相比,敏感性为91.67%,特异性为77.14%,Kappa值为0.577。结论对于感染性眼内炎,PCR联合直接基因测序技术是一种快速﹑有效的临床检查技术。
Objective To explore the feasibility and effectiveness of using polymerase chain reaction (PCR) in combination with direct gene sequencing to detect infectious endophthalmitis and to seek a new detection method for the rapid diagnosis of infectious endophthalmitis. Methods Forty-seven patients (47 eyes) with infectious endophthalmitis surgically treated in our hospital from October 2005 to October 2006 were examined by PCR combined with direct gene sequencing and were compared with bacterial smear, bacterial culture, fungal culture, anaerobic Comparison of bacterial culture test results. Results PCR combined with gene diagnostic methods to check the time less time consuming than the traditional method. PCR positive rate of pathogen detection was high, 25 cases, 25 cases were positive by direct gene sequencing of pathogens in 19 cases. Compared with the pathogen culture, the sensitivity was 91.67%, the specificity was 77.14% and the Kappa value was 0.577. Conclusion Infectious endophthalmitis, PCR combined with direct gene sequencing is a rapid and effective clinical examination technique.
