探讨胃癌患者外周血癌细胞的检测及其意义。方法:以CK19作指标,用巢式RT-PCR方法检测42例胃癌患者术前外周血中癌细胞。结果:健康人外周血中不能检测出CK19mRNA表达,用巢式RT-PCR方法检测外周血中胃癌细胞的敏感性可达1/106;42例胃癌病人外周血中检测出CK19mRNA阳性表达13例,阳性率30.9%,阳性结果与肿瘤大小、浸润深度和肿瘤分期密切相关。结论:巢式RT-PCR方法检测胃癌外周血癌细胞具有高度的特异性和敏感性,检测外周血癌细胞有助于判断肿瘤的进展程度。
To investigate the detection of peripheral blood cancer cells in patients with gastric cancer and its significance. Methods: Cytokeratin 19 (CK19) was used as a marker to detect the preoperative cancer cells in 42 patients with gastric cancer by nested RT-PCR. Results: CK19mRNA was not detected in the peripheral blood of healthy people. The sensitivity of nested RT-PCR to detect gastric cancer cells in peripheral blood was up to 1/106. The positive expression of CK19mRNA in peripheral blood of 42 patients with gastric cancer was detected in 13 cases, Positive rate of 30.9%, positive results and tumor size, depth of invasion and tumor stage are closely related. Conclusion: The detection of peripheral blood cancer cells by nested reverse transcriptase-polymerase chain reaction (RT-PCR) is highly specific and sensitive. Detecting peripheral blood cancer cells can be helpful in judging the tumor progression.
