运用高效液相色谱法,对藏药小叶莲中3种木脂素(鬼臼毒素、去氧鬼臼毒素和4’-去甲去氧鬼臼毒素)和5种黄酮类成分(8-异戊烯基山柰酚、槲皮素、山柰酚、8,2’-二异戊烯基槲皮素3-甲醚、8-异戊烯基槲皮素)进行含量测定,采用Agilent Zorbax SB-C18色谱柱,乙腈–0.05%的磷酸水溶液作为流动相进行梯度洗脱;流速1 m L/min;柱温35°C;检测波长为290 nm、370 nm。该方法呈现良好的稳定性:日内、日间精密度的RSD值分别小于3%、4%。各对照品的线性及相关性良好(R2≥0.9992),加样回收率测定值在98.29%–101.60%之间。采用上述方法测定了12份小叶莲(桃儿七果实)和5份桃儿七根及根茎中8种有效成分的含量。结果表明:8种被测成分在所测样品中的含量差异明显(总含量值从16.90 mg/g到55.68 mg/g之间不等),同时发现异戊烯基黄酮类成分是小叶莲中的代表性成分,可用于小叶莲的质量控制和鉴别研究。
The effects of three kinds of lignan (podophyllotoxin, deoxypodophyllotoxin and 4’-demethoxypodophyllotoxin) and five flavonoid components (8-different Pentenyl kaempferol, quercetin, kaempferol, 8,2’-diisoprenyl quercetin 3-methyl ether, 8-prenyl quercetin) were determined by Agilent Zorbax SB C18 column, gradient elution with acetonitrile-0.05% phosphoric acid in water as mobile phase; flow rate 1 m L / min; column temperature 35 ° C; detection wavelength 290 nm, 370 nm. The method showed good stability: RSD values of intra-day and inter-day precision were less than 3% and 4%, respectively. The linearity and correlation of each reference substance was good (R2≥0.9992). The recoveries of samples were between 98.29% and 101.60%. Using the above method, the contents of 8 active ingredients in 12 small lotus (peach seven fruits) and 5 peach seven and rhizomes were determined. The results showed that the contents of 8 tested components were significantly different among the tested samples (the total content ranged from 16.90 mg / g to 55.68 mg / g), and the isopentenyl flavonoids The representative components, can be used for quality control and identification of leaf lotus.
