目的:发现具有类似神经生长因子(nerve growth factor,NGF)活性的小分子化合物。方法:通过生物活性实验筛选,发现白木耳的甲醇浸提物具有显著诱导PC12细胞发生神经突起伸长的作用。在生物活性鉴定实验的引导下,通过多次柱色谱分离,HPLC纯化,得到一纯净的活性化合物1,利用波谱分析法确定了其化学结构。结果:通过与文献的氢谱、碳谱、质谱对比,确定该活性分子属麦角甾醇苷类化合物,是3-O-β-D-glucopyranosyl-22E,24R-5α,8α-epidioxyergosta-6,22-diene。该化合物在3μmol.L-1浓度下具有显著诱导PC12细胞发生神经突起伸长的活性。结论:来源于白木耳的麦角甾醇苷类的化合物1具有显著的类似神经生长因子NGF的活性。
Objective: To find small molecule compounds with similar activity of nerve growth factor (NGF). Methods: The biological activity of experimental screening, found that white fungus methanol extract significantly induced PC12 cell neurite outgrowth role. Under the guidance of bioassay, a series of column chromatography and HPLC were purified to obtain a pure active compound 1, whose chemical structure was confirmed by spectral analysis. Results: By contrast with the literature of the hydrogen, carbon and mass spectra, it is confirmed that the active molecule belongs to the class of 3-O-β-D-glucopyranosyl-22E, 24R-5α, 8α-epidioxyergosta-6,22 -diene. The compound has a remarkable activity of inducing neurite outgrowth in PC12 cells at a concentration of 3 μmol·L-1. Conclusion: Compound 1 of the ergosterol glycosides derived from Agrocybe aegerus has significant NGF-like activity.
