目的:观察脾虚大鼠胃粘膜TFF1mRNA与蛋白表达变化及补中益气汤对其表达的影响,并探讨相关作用机制。方法:SD大鼠随机分为空白对照、脾虚损伤、补中益气汤、非脾虚损伤组。脾虚损伤组、补中益气汤组大鼠给予100%大黄水煎剂灌胃,一天2次,连续10天。第11天补中益气汤组给予补中益气汤灌胃治疗7天。第18天,除空白对照组外,另三组灌服消炎痛溶液造成胃粘膜损伤,采用逆转录PCR(RT-PCR)检测TFF1mRNA,采用双抗体两步夹心酶联免疫吸附法(ELISA)检测TFF1蛋白表达的变化。结果:与非脾虚损伤组比较,脾虚损伤组大鼠胃粘膜TFF1mRNA、TFF1蛋白表达明显降低;与脾虚损伤组比较,补中益气汤组大鼠胃粘膜TFF1mRNA、蛋白明显上调。结论:脾虚大鼠消炎痛攻击后胃粘膜TFF1mRNA、蛋白表达显著下调,提示脾虚大鼠胃粘膜防御能力减弱,补中益气汤可提高脾虚大鼠消炎痛攻击后胃粘膜TFF1mRNA与蛋白的表达,这可能是补中益气汤保护胃粘膜,降低脾虚大鼠胃粘膜易损性的机理之一。
Objective: To observe the changes of TFF1 mRNA and protein expression in gastric mucosa of rats with spleen deficiency and the effect of Buzhong Yiqi Decoction on its expression, and to explore the related mechanism. Methods: SD rats were randomly divided into blank control, spleen deficiency, Buzhong Yiqi Decoction, non-spleen deficiency group. Rats in the spleen deficiency group and the Buzhong Yiqi Decoction group were given 100% rhubarb water decoction orally, twice a day for 10 consecutive days. On the 11th day, Buzhong Yiqi Decoction group was given Buzhong Yiqi Decoction for 7 days. On the 18th day, in addition to the blank control group, the other three groups were infused with indomethacin solution to cause gastric mucosal injury. TFF1 mRNA was detected by reverse transcription-PCR (RT-PCR) and detected by double antibody two-step sandwich enzyme-linked immunosorbent assay (ELISA). Changes in TFF1 protein expression. Results:Compared with non-spleen deficiency group, the expression of TFF1 mRNA and TFF1 protein in the gastric mucosa of the spleen deficiency group was significantly decreased. Compared with the spleen deficiency group, the TFF1 mRNA and protein of the gastric mucosa in the Buzhong Yiqi Decoction group were significantly increased. Conclusion: The expression of TFF1 mRNA and protein in the gastric mucosa was significantly down-regulated after indomethacin challenge in rats with spleen deficiency, suggesting that the gastric mucosal defense ability of spleen deficiency rats was weakened. Buzhong Yiqi Decoction could increase the expression of TFF1 mRNA and protein in gastric mucosa after indomethacin challenge in rats with spleen deficiency. This may be one of the mechanisms by which Buzhong Yiqi Decoction protects gastric mucosa and reduces the vulnerability of gastric mucosa in rats with spleen deficiency.
