本文采用整体犬急性心肌缺血-再灌注模型以探讨穿心莲提取液减轻钙超负荷的机制。结扎冠脉左前降支上1/3处90min后再灌注60min较持续结扎150min,缺血中心区心肌细胞内Ca2+增加(P<0.05)、Na+明显增加(P<0.01),心肌细胞膜Ca2+-ATPase活性明显降低(P<0.01),心肌组织MDA含量显著增高(P<0.01)。而在再灌注前45min静脉给予穿心莲提取液,则见其缺血中心区心肌细胞内Ca2+降低(P<0.05)、Na+明显降低(P<0.01),心肌细胞膜Na+-K+-ATPase、Ca2+-ATPase活性明显增加(P<0.01)。心肌组织MDA含量显著降低(P<0.01)。说明穿心莲提取液减轻心肌缺血-再灌注过程中钙超负荷与减轻氧自由基危害、保护心肌细胞膜ATPase活性、降低钠超负荷有关。
In this paper, the whole canine model of acute myocardial ischemia-reperfusion was used to investigate the mechanism of andrographis paniculata extract on reducing calcium overload. And 45 minutes before reperfusion perfusion of Andrographis paniculata extract, then the ischemic center of myocardial cell Ca2 + decreased (P <0.05), Na decreased significantly (P <0.01), myocardial cell membrane Na + -K + -ATPase , Ca2 + -ATPase activity increased significantly (P <0.01). Myocardial MDA content was significantly lower (P <0.01). It indicates that the extract of Andrographis paniculata can relieve myocardial ischemia-reperfusion process, calcium overload and reduce the harm of oxygen free radicals, protect myocardial cell membrane ATPase activity, reduce sodium overload.
