目的 探讨p16基因变异及其产物表达与食管癌生物学行为的关系。方法 应用PCR SSCP和PCR方法检测了林县食管癌标本中p16 CDKN2 基因 1、2外显子的突变和缺失状况。并用免疫组化方法检测其产物表达情况。结果 在 47例标本中 ,3例出现了突变 ,其中第 1外显子 1例 ,第 2外显子 2例 ,突变率为 6 4% ( 3 47)。有 12例出现纯合性缺失 ,缺失率为 2 5 5 % ,其总变异率为 31 9%。免疫组化染色显示 ,47例食管癌组织中 2 0例出现 p16蛋白的表达 ,阳性率为 48 8%。结论 结论 p16基因变异可能在食管癌的发生中发挥一定的作用。p16蛋白的表达与食管癌的组织学分级之间有一定的关系 ,与预后无明显相关性
Objective To investigate the relationship between the mutation of p16 gene and the expression of p16 gene and the biological behavior of esophageal cancer. Methods The mutations and deletions of exon 1 and exon 1 of p16 CDKN2 gene in esophageal cancer specimens of Linxian were detected by PCR SSCP and PCR. Immunohistochemistry was used to detect the product expression. Results Of the 47 specimens, 3 had mutations, of which 1 was exon 1 and 2 was exon 2, with a mutation rate of 64% (3 47). There were 12 cases of homozygous deletion, the deletion rate was 25.5%, the total mutation rate was 31 9%. Immunohistochemical staining showed that p16 protein was found in 20 of 47 esophageal cancer tissues, with a positive rate of 48.8%. Conclusion Conclusion The mutation of p16 gene may play a role in the carcinogenesis of esophageal cancer. There is a certain relationship between the expression of p16 protein and the histological grade of esophageal cancer, and no significant correlation with prognosis
