目的探讨CD28单抗对细胞增殖效果及细胞毒作用,分析CD28单抗对CD3-AK肿瘤杀伤活性的调节作用。方法制备CD28单克隆抗体,诱导CD3-AK细胞,用倒置显微镜观察细胞数量和形态,用MTT法测定CD3-AK细胞对骨髓瘤细胞的抑制率。结果CD3-AK细胞加入CD28单抗,细胞数量明显增多(t=10.6,P<0.05);对肿瘤细胞的杀伤活性明显增高。结论CD28单抗与CD3单抗联合,具有较强的协同作用,能有效提高CD3-AK的肿瘤杀伤活性。
Objective To investigate the effect of CD28 monoclonal antibody on cell proliferation and cytotoxicity and to study the regulatory effect of CD28 monoclonal antibody on CD3-AK tumor activity. Methods CD28 monoclonal antibody was used to induce CD3-AK cells. The number and morphology of CD3-AK cells were observed by inverted microscope. The inhibitory rate of CD3-AK cells on myeloma cells was determined by MTT assay. Results CD3-AK cells with CD28 monoclonal antibody significantly increased the number of cells (t = 10.6, P <0.05), and significantly increased the killing activity of tumor cells. Conclusion The combination of CD28 McAb and CD3 monoclonal antibody has a strong synergistic effect and can effectively increase the tumor-killing activity of CD3-AK.
