目的:建立HPLC法同时测定半合成紫杉醇中的10-去乙酰巴卡亭Ⅲ、巴卡亭Ⅲ、7-三氯乙氧羰酰巴卡亭Ⅲ、13-O-[(4S,5R)-2-(4-甲氧基苯基)-N-苯甲酰基-4-苯基-噁唑烷基]-7-三氯乙氧羰酰巴卡亭Ⅲ和紫杉醇的含量。方法:用Angilent ODS C18分析柱(4.6 mm×150 mm,5μm),甲醇-乙腈-0.1%乙酸水溶液线性梯度洗脱:0~8.5 min(24:31:45),8.5~38.5 min(57:43:0),38.5~60 min(24:31:45);流速0.8 mL.min-1,检测波长227 nm。结果:该测定方法回收率高,成分的分离度好。结论:该法简便快速,重现性好,适用于半合成紫杉醇的质量控制。
OBJECTIVE: To establish a HPLC method for the simultaneous determination of 10-deacetylbaccatin III, baccatin III, 7-trichloroethoxycarbonyl baccatin III, 13-O - [(4S, 5R) - 2- (4-methoxyphenyl) -N-benzoyl-4-phenyl-oxazolidinyl] -7-trichloroethoxycarbonyl baccatin III and paclitaxel. Methods: A linear gradient of methanol-acetonitrile-0.1% aqueous acetic acid was performed on a Angilent ODS C18 column (4.6 mm × 150 mm, 5 μm) at 0 to 8.5 min (24:31:45) and 8.5 to 38.5 min 43: 0), 38.5-60 min (24:31:45), flow rate of 0.8 mL.min-1 and detection wavelength of 227 nm. Results: The method has high recovery and good separation of components. Conclusion: The method is simple and rapid, reproducible, suitable for semi-synthetic paclitaxel quality control.
