目的:研究褪黑素(MT)对谷氨酸所致脑组织氧化损伤模型小鼠的对抗作用。方法:将小鼠随机分为空白组、模型组、MT(高、低剂量)组,MT组预先经腹腔注射给药7d ;经侧脑室注射谷氨酸造模,测定各组小鼠脑组织中的丙二醛(MDA)、谷胱甘肽(GSH)含量及谷胱甘肽过氧化物酶(GSH -PX)活性。结果:模型组小鼠脑组织MDA含量显著升高(P<0 .01) ,GSH含量显著降低(P<0 .01) ;与模型组比较,MT组MDA含量降低,GSH含量升高,GSH -PX活性增强。结论:MT可通过抗氧化及增强抗氧化酶活性抑制谷氨酸的神经毒性,发挥抗衰老作用。
Objective: To study the antagonistic effect of melatonin (MT) on glutamate-induced oxidative injury in brain tissue of mice. Methods: The mice were randomly divided into blank group, model group, MT group (high dose and low dose), MT group was pre-treated by intraperitoneal injection for 7 days. After intracerebroventricular injection of glutamate, Malondialdehyde (MDA), glutathione (GSH) and glutathione peroxidase (GSH-PX) activity were measured. Results: Compared with the model group, the content of MDA and the content of GSH in model group were significantly increased (P <0.01, P <0.01) -PX activity increased. CONCLUSION: MT can inhibit the neurotoxicity of glutamate through anti-oxidation and anti-oxidative enzyme activity and exert anti-aging effects.
