目的:探讨野菊花对压力负荷性大鼠心室重构及神经内分泌因子的影响。方法:腹主动脉不完全结扎法(AAB)制作大鼠心肌肥厚、心室重构模型。35d药物干预后,测量大鼠收缩压(SBP)、舒张压(DBP)、平均动脉压(MAP);称重法测定心脏指数(HW/BW、LVW/BW);放免法测定心肌血管紧张素Ⅱ(AngⅡ),血清醛固酮(ALD)、肿瘤坏死因子α(TNF-α)的水平;心肌样本碱水法测定羟脯氨酸(Hyp)的含量。结果:模型组SBP、DBP、MAP、HW/BW、LVW/BW、AngⅡ、ALD、TNF-α及Hyp含量升高(P<0.05)。野菊花改善心肌肥厚指数,降低AngⅡ、ALD、TNF-α及Hyp含量作用显著(P<0.05)。结论:野菊花通过抑制压力负荷性心室重构大鼠RAAS和交感神经系统活性,减少神经内分泌因子AngⅡ、TNF-α、ALD生成而抑制心肌肥大、心室重构,这可能是野菊花直接保护心肌,抑制心肌细胞纤维化的作用机制之一。
Objective: To investigate the effect of wild chrysanthemum on ventricular remodeling and neuroendocrine factors in pressure overload rats. Methods: Cardiac hypertrophy and ventricular remodeling were induced by abdominal aorta incomplete ligation (AAB). After 35 days of drug intervention, systolic blood pressure (SBP), diastolic blood pressure (DBP) and mean arterial pressure (MAP) were measured; cardiac index (HW/BW, LVW/BW) was measured by weighing method; and myocardial angiotensin was measured by radioimmunoassay. II (Ang II), serum aldosterone (ALD), tumor necrosis factor α (TNF-α) levels; myocardial samples alkaline water method for the determination of hydroxyproline (Hyp) content. Results: The levels of SBP, DBP, MAP, HW/BW, LVW/BW, AngII, ALD, TNF-α and Hyp were increased in the model group (P<0.05). Wild chrysanthemum improved myocardial hypertrophy index, and decreased AngII, ALD, TNF-α and Hyp content significantly (P<0.05). Conclusion: Wild Chrysanthemum inhibits the activity of RAAS and sympathetic nervous system in rats with pressure-induced ventricular remodeling, reduces the production of neuroendocrine factors AngII, TNF-α and ALD and inhibits cardiac hypertrophy and ventricular remodeling. , One of the mechanisms that inhibit myocardial fibrosis.
