为制备针对柱状黄杆菌的单克隆抗体(McAb),利用颗粒性抗原免疫BALB/c小鼠,将其脾细胞与小鼠骨髓瘤SP2/0细胞融合。经过有限稀释法筛选,得到2株稳定分泌抗柱状黄杆菌McAb的杂交瘤细胞株(2B5和4G7)。其分泌抗体亚类属于IgG1,轻链属于κ链。制备的McAb与迟钝爱德华氏菌、大肠杆菌、嗜水气单胞菌、鳗弧菌、溶藻弧菌、副溶血弧菌、哈维氏弧菌等水产病原菌均无交叉反应。表明首次成功建立了抗柱状黄杆菌McAb的制备方法,为柱状黄杆菌的快速检测奠定了基础。
To prepare a monoclonal antibody (McAb) directed against Flavobacterium columnarum, BALB / c mice were immunized with particulate antigen and their spleen cells were fused with mouse myeloma SP2 / 0 cells. After screening by limiting dilution method, two hybridoma cell strains (2B5 and 4G7) were successfully secreted McAb against P. cylindromator. The secretory antibody subclass belongs to IgG1 and the light chain belongs to the kappa chain. The prepared McAbs had no cross reaction with the pathogenic bacteria of Edwardsiella spp., Escherichia coli, Aeromonas hydrophila, Vibrio anguillarum, Vibrio alginolyticus, Vibrio parahaemolyticus and Vibrio harveyi. The results showed that the first method for the preparation of McAb against Pseudomonas cylindricalis was successfully established, which lays the foundation for the rapid detection of the bacterium Flavobacterium columnarum.
