目的 的观察华支睾吸虫脂肪酸结合蛋白(Clonorchis sinensis fatty acid binding protein,CsFABP)作用后小鼠巨噬细胞RAW264.7表面分子表达、细胞吞噬活性及NO、TNF-a分泌情况,探讨CsFABP对巨噬细胞免疫功能的影响. 方法 以LPS为阳性对照,分别用CsFABP和LPS+ CsFABP处理RAW264.7细胞,采用流式细胞术检测细胞表面抗原CD80和MHCII,采用中性红法检测RAW264.7细胞的吞噬活性,采用4-氨基安替吡啉法检测RAW264.7细胞的酸性磷酸酶含量,采用Griess法检测细胞培养上清中NO分泌水平,采用ELISA检测TNF-α分泌水平. 结果 与对照组相比,CsFABP处理24 h后巨噬细胞吞噬中性红的能力增强(P<0.05),酸性磷酸酶的含量增加(P<0.01),NO和TNF-α分泌增加(P<0.05),但巨噬细胞表面抗原CD80和MHCII的表达无明显变化(P>0.05).当CsFABP与LPS联合作用时能显著降低LPS引起的细胞表面抗原CD80和MHCII的升高(P<0.05),降低巨噬细胞的吞噬活性(P<0.05). 结论 CsFABP可影响巨噬细胞RAW264.7的吞噬活性,增加巨噬细胞NO、TNF-α的分泌,对巨噬细胞免疫功能起调节作用.“,”Objective To investigate the effects of Clonorchis sinensis fatty acid-binding protein (CsFABP) on the expression of surface molecules,phagocytic activity,and production of NO and TNF-α by RAW264.7 macrophages and to examine the mechanism by which CsFABP affects the immune function of macrophages.Methods Macrophages were treated with LPS,CsFABP,or LPS+ CsFABP for 24 h,and cell surface molecules such as CD80 and MHCII were detected with flow cytometry.The phagocytic activity of macrophages was indicated by neutral red,the ability of acid phosphatase activity was measured with 4-aminoantipyrine (4-AAP),NO production was detected with a Griess reagent system,and TNF-α in culture supernatant was measured with ELISA.Results When the macrophages were stimulated by CsFABP for 24 h,the phagocytic activity of RAW264.7 cells stimulated with CsFABP alone was significantly greater than that of the control (P<0.05),the level of acid phosphatase in macrophages also increased (P<0.01),and the levels of NO and TNF-α increased significantly (P<0.05).In contrast,there were no significant effects on the expression of CD80 and MHCII on the surface of RAW264.7 macrophages stimulated with CsFABP for 24 h.When CsFABP was combined with LPS,the increase in the levels of CD80 and MHCII induced by LPS decreased significantly (P<0.05) and the phagocytic activity of macrophages decreased (P<0.05).Conclusion CsFABP may play a regulatory role in the function of macrophages during C.sinensis infection.
