目的对比计算机图像分析与人工计数阳性细胞方法在检测GFAP阳性细胞中应用,探讨计算机图像分析代替人工计数的可行性及优缺点。方法应用正常组、癫痫组以及灵芝孢子粉组三组大鼠切25μ厚冰冻切片,常规GFAP免疫组化染色。人工计数GFAP阳性细胞,同时用Image-Pro Plus分析图片的积分光密度,对比各组间差异以及两种方法之间的相关性。结果两种方法都能检测出各组间存在差别,结果具有相关性。结论用人工计数方法和计算机图像处理结果具有正相关关系,其得出的结论也一致,在某些可代替人工技术方法用于免疫组化分析。
Objective To compare the application of computer image analysis and manual counting of positive cells in the detection of GFAP positive cells and to explore the feasibility and advantages and disadvantages of computer image analysis instead of manual counting. Methods The rats in normal group, epilepsy group and Ganoderma lucidum spore powder group were cut into 25μ thick frozen sections and routine GFAP immunohistochemical staining. The GFAP positive cells were counted manually. At the same time, the integral optical density of the images was analyzed by Image-Pro Plus. The differences between the groups and the correlation between the two methods were compared. Results Both methods detected differences between the groups and the results were correlated. Conclusions There is a positive correlation between the artificial counting method and the computer image processing result. The conclusion reached is also consistent. Some alternative artificial techniques are used for immunohistochemistry analysis.
