目的:探讨他莫昔芬(tamoxifen,TAM)联合多柔比星(doxorubicin,ADR)对人骨肉瘤MG-63细胞生长的抑制作用,比较二者联合作用的疗效是否优于单药。方法:采用RT-PCR法检测人骨肉瘤MG-63细胞的雌激素受体(estrogen receptor,ER)mRNA表达。应用倒置显微镜观察TAM和ADR单药及两药联合作用下各组细胞的形态学变化。MTT法检测单药及两药联合作用下各组细胞的生长抑制情况。结果:人骨肉瘤MG-63细胞表达ER。细胞形态学观察发现各组细胞用药后,与对照组相比均可见生长抑制变化。MTT法检测显示TAM及ADR均可在体外抑制人骨肉瘤MG-63细胞的增殖,两药联合组细胞增殖抑制率高于2个同等浓度单药组,甚至高于2倍浓度单独用药组,差异具有统计学意义(P<0.05)。结论:人骨肉瘤MG-63细胞表达ER,ADR及TAM以及两药联用均可抑制细胞增殖,两药联合应用组细胞抑制率高于单药组。推测其机制可能是由于TAM能增加人骨肉瘤MG-63细胞对化疗药物ADR的敏感性所致。
OBJECTIVE: To investigate the inhibitory effect of tamoxifen (TAM) and doxorubicin (ADR) on the growth of human osteosarcoma MG-63 cells, and to compare whether the combined effect of tamoxifen (TAM) and monotherapy is better than that of monotherapy. Methods: The mRNA expression of estrogen receptor (ER) in human osteosarcoma MG-63 cells was detected by RT-PCR. Inverted microscope was used to observe the morphological changes of TAM and ADR single cells and the combination of the two drugs. MTT assay was used to detect the growth inhibition of each group of cells under the combined action of single drug and two drugs. Results: Human osteosarcoma MG-63 cells expressed ER. Morphological observation showed that after administration of each group of cells, compared with the control group, growth inhibition was observed. MTT assay showed that both TAM and ADR could inhibit the proliferation of human osteosarcoma MG-63 cells in vitro. The inhibitory rate of cell proliferation in combination group was higher than that in two equal concentration groups or even higher than twice concentration group Statistically significant (P <0.05). Conclusion: The expression of ER, ADR and TAM in human osteosarcoma MG-63 cells and the combination of the two drugs can inhibit the cell proliferation. The inhibitory rate of the combination of the two drugs is higher than that of the single drug group. It is speculated that the mechanism may be due to TAM can increase the sensitivity of human osteosarcoma MG-63 cells to chemotherapy ADR.
