目的探讨沙利度胺(THD)对人胰腺癌细胞SW1990裸鼠皮下移植瘤生长的影响。方法建立20只人胰腺癌裸鼠皮下移植瘤模型后均分为4组,分别用THD 200mg·kg-1·d-1(A组)、吉西他滨(GEM)50mg·kg-1·d-1(B组)、THD+GEM(C组)和生理盐水(D组)干预。28d后处死裸鼠,测量各组肿瘤重量,计算抑瘤率。结果 A、B和C组移植瘤重量分别为(1.013±0.096)g、(0.864±0.112)g和(0.512±0.091)g,明显轻于D组的(1.412±0.138)g(P<0.05)。A、B和C组抑瘤率分别为28.3%、38.8%和63.7%,C组抑瘤率高于A、B组(P<0.05)。结论THD对胰腺癌裸鼠皮下移植瘤的生长具有抑制作用,与GEM联合应用对肿瘤抑制作用更强。
Objective To investigate the effects of thalidomide (THD) on the growth of subcutaneous xenografts in human pancreatic cancer cell line SW1990. Methods Twenty adult human pancreatic cancer xenografts were divided into 4 groups: THD 200 mg · kg -1 · d -1 (group A), Gemcitabine (GEM) 50 mg · kg -1 · d -1 (Group B), THD + GEM (group C) and saline (group D). After 28 days, the nude mice were sacrificed and the tumor weight of each group was measured to calculate the tumor inhibition rate. Results The tumor weights in groups A, B and C were (1.013 ± 0.096) g, (0.864 ± 0.112) g and (0.512 ± 0.091) g respectively, which were significantly lower than those in group D (1.412 ± 0.138 g) (P <0.05) . The inhibition rate of tumor in groups A, B and C were 28.3%, 38.8% and 63.7%, respectively. The inhibition rate of tumor in group C was higher than that in group A and B (P <0.05). Conclusion THD can inhibit the growth of subcutaneously transplanted pancreatic cancer in nude mice, and its combination with GEM can inhibit tumor growth more strongly.
