目的分析使用雄黄混悬液对卵巢癌SKOV3细胞增殖抑制和诱导凋亡机制的影响。方法体外培养的SKOV3细胞分别与不同浓度的纳米雄黄混悬液孵育24、48、72和96 h,应用MTT法检测细胞增殖抑制率,采用原位末端标记法(TUNEL)对细胞凋亡形态特征进行观察,分析细胞周期变化,应用Western-blot检测SKOV3细胞Caspase-3蛋白的表达。结果不同浓度纳米雄黄混悬液对SKOV3细胞均有增殖抑制作用,且呈浓度和时间依赖性及周期特异性生长抑制作用;能明显增强Caspase-3蛋白的表达水平。结论纳米雄黄混悬液可在体外抑制SKOV3细胞增殖并诱导凋亡。其机制可能与诱导细胞发生G1期阻滞并激活Caspase途径有关。
Objective To analyze the effect of realgar suspension on proliferation and apoptosis-inducing mechanism of ovarian cancer SKOV3 cells. Methods SKOV3 cells cultured in vitro were incubated with different concentration of realgar suspension for 24, 48, 72 and 96 h respectively. The inhibition rate of cell proliferation was detected by MTT assay. The morphological changes of apoptotic cells were detected by TUNEL method. The changes of cell cycle were analyzed. The expression of Caspase-3 protein in SKOV3 cells was detected by Western-blot. Results The suspension of nano-realgar at different concentrations could inhibit the proliferation of SKOV3 cells in a concentration-dependent and time-dependent manner and inhibited the growth of SKOV3 cells. The expression of Caspase-3 protein was significantly enhanced. Conclusion Nano realgar suspension can inhibit SKOV3 cell proliferation and induce apoptosis in vitro. The mechanism may be related to induction of G1 arrest and activation of Caspase pathway.
