目的探讨丹参川芎嗪注射液对缺氧条件下脑基底平滑肌细胞的保护作用,并比较丹参素、川芎嗪、丹参川芎嗪注射液三者的药效差异。方法用Na_2S_2O_4与细胞共孵育,建立大鼠脑基底平滑肌细胞缺氧/复氧损伤模型。对照组换上新鲜培养基;模型组给予缺氧液;丹参川芎嗪低、中、高浓度组分别给予0.01,0.1,1μmol·L~(-1)丹参川芎嗪注射液孵育6 h;丹参素组给予0.1μmol·L~(-1)丹参素孵育6 h;川芎嗪组给予0.1μmol·L~(-1)川芎嗪孵育6 h;丹参素+川芎嗪组给予0.1μmol·L~(-1)丹参素+0.1μmol·L~(-1)川芎嗪孵育6 h。观察脑基底平滑肌细胞一般形态学及凋亡相关形态学的改变、检测脑基底平滑肌细胞存活率、脑基底平滑肌细胞外液乳酸脱氢酶活性、以及Bcl-2/Bax的表达。结果与模型组相比较,给药组脑基底平滑肌细胞的存活率上升(P<0.05或0.01),Bcl-2/Bax的比值升高(P<0.05),降低了低氧损伤引起的细胞外液乳酸脱氢酶的升高(P<0.05)。结论丹参川芎嗪注射液对缺氧造成的脑基底平滑肌细胞损伤具有保护作用,其保护作用优于丹参素、川芎嗪。
Objective To investigate the protective effect of Salvia miltiorrhizae ligustrazine injection on cerebral basal smooth muscle cells under hypoxic conditions and to compare the pharmacodynamic effects of Danshensu, ligustrazine and Salviae miltiorrhizae ligustrazine injection. Methods Na_2S_2O_4 was co-incubated with cells to establish an anoxia / reoxygenation injury model of rat basal smooth muscle cells. The control group was replaced with fresh medium; the model group was given hypoxic solution; the low, middle and high concentration of Ligustrazine Ligustrazine group were respectively given 0.01, 0.1, 1μmol·L -1 Salviae Miltiorrhizae ligustrazine injection for 6 h; Danshensu Group was given 0.1μmol·L -1 Danshensu for 6 hours, ligustrazine group was given 0.1μmol·L -1 ligustrazine for 6 hours, Danshensu + Ligustrazine group was given 0.1μmol·L ~ (-1) 1) Danshensu + 0.1 μmol·L -1 ligustrazine for 6 h. The changes of morphology and apoptosis related morphological changes of basal smooth muscle cells were observed. The survival rate of basal smooth muscle cells, extracellular fluid lactate dehydrogenase and the expression of Bcl-2 / Bax in basal smooth muscle cells were measured. Results Compared with the model group, the viability of basal smooth muscle cells increased (P <0.05 or 0.01) and the ratio of Bcl-2 / Bax increased (P <0.05), and the extracellular load Lactate dehydrogenase increased (P <0.05). Conclusion Salviae miltiorrhizae ligustrazine injection has a protective effect on hypoxic-induced injury of basal smooth muscle cells and its protective effect is better than that of Danshensu and ligustrazine.
