目的:观察中药复方芪丹地黄颗粒对2型糖尿病肾病(type 2 diabetic nephropathy)db/db小鼠肾脏功能的保护作用,为芪丹地黄颗粒治疗糖尿病肾病提供实验依据。方法:取8周龄db/db小鼠12只,根据空腹血糖(fasting blood glucose,FBG)分为模型组及芪丹地黄颗粒组,每组6只,另取6只db/+小鼠为正常对照组。正常对照组及模型组正常进食进水,给药组予芪丹地黄含药饲料喂养。治疗共16周,于第4、8、12、16周末检测各组空腹血糖,并在第16周末收集小鼠24 h尿液测定尿蛋白排泄率(24 hour urinary albumin excretion rate,24 h-UAER),随后处死小鼠,取血清测定肌酐(serum creatinine,SCr)、尿素(blood urea nitrogen,BUN)、血脂,取肾脏进行病理形态学检查及透射电镜检测。结果:实验16周后模型组小鼠FBG、BUN、SCr、24 h-UAER、胆固醇(cholesterol,TC)、甘油三酯(triglyceride,TG)、低密度脂蛋白(low density Lipoprotein,LDL)较正常组明显升高(P<0.05),给药组FBG、BUN、SCr、24 h-UAER、TG较模型组显著降低(P<0.05),TC及LDL未见统计学差异。组织病理学见模型组肾小球内皮细胞和系膜细胞数量减少,糖原沉积增加,纤维细胞增生,透射电镜见模型组基底膜明显增厚,足突减少,给药治疗后上述病变明显减轻。结论:芪丹地黄颗粒明显改善2型糖尿病肾病小鼠的肾损害程度,保护肾功能。
OBJECTIVE: To observe the protective effect of Qi Dan Di Huang granule on kidney function of type 2 diabetic nephropathy db / db mice, and to provide experimental evidence for the treatment of diabetic nephropathy with Qi Dan Di Huang granules. Methods: Twelve db / db mice aged 8 weeks were divided into model group and qidandihuang granule group according to fasting blood glucose (FBG) group, with 6 mice in each group and 6 db / + mice Normal control group. The normal control group and the model group were fed with normal water, while the administration group was fed with the medicated feed of Qidan Dihuang. After a total of 16 weeks of treatment, fasting blood glucose was measured at the end of the 4th, 8th, 12th and 16th weeks. 24-hour urinary albumin excretion rate (24 h-UAER) The mice were then sacrificed and serum creatinine (SCr), blood urea nitrogen (BUN) and blood lipids were determined. Pathological examination and transmission electron microscopy were performed on the kidneys. Results: After 16 weeks, FBG, BUN, SCr, 24 h-UAER, cholesterol (TC), triglyceride (TG) and low density lipoprotein (LDL) (P <0.05). The FBG, BUN, SCr, 24 h-UAER, TG in the treatment group were significantly lower than those in the model group (P <0.05), and there was no significant difference between the TC and LDL groups. Histopathology model group, the number of glomerular endothelial cells and mesangial cells decreased, increased glycogen deposition, fibroblast proliferation, the model group by transmission electron microscopy was significantly thicker basement membrane, foot process reduced, after treatment, the above lesions were significantly reduced . Conclusion: Qidan Dihuang Granules can obviously improve the degree of renal damage in type 2 diabetic nephropathy mice and protect renal function.
