目的:研究激活淋巴细胞和未激活淋巴细胞的端粒酶逆转录酶(human telomerase reverse transcriptase,hTERT)的表达情况。方法:运用定量RT-PCR法检测分别代表激活淋巴细胞和未激活淋巴细胞的43枚良性结直肠疾病的炎性系膜淋巴结和31例正常自愿者的外周血单核细胞hTERT的表达水平。另外,作为阳性和阴性对照,还检测了59对结直肠癌肿瘤组织/正常黏膜组织的hTERT表达水平。结果:炎性系膜淋巴结的hTERT表达水平比正常人外周血单核细胞的高,但没有统计学意义(P=0.07)。肿瘤组织的hTERT表达水平明显比正常组织的高(P=0.001)。结论:淋巴细胞无论激活与否,hTERT表达水平没有显著改变,与正常组织也无明显差异。
Objective: To investigate the expression of human telomerase reverse transcriptase (hTERT) in activated lymphocytes and inactivated lymphocytes. Methods: The expression of hTERT in peripheral blood mononuclear cells from inflammatory mesangial lymph nodes of 43 patients with benign colorectal diseases and 31 normal volunteers were detected by quantitative RT-PCR. In addition, hTERT expression levels in 59 pairs of colorectal cancer tissues / normal mucosa were also examined as positive and negative controls. Results: The expression of hTERT in inflammatory mesangial lymph nodes was higher than that of normal human peripheral blood mononuclear cells, but there was no statistical significance (P = 0.07). The expression of hTERT in tumor tissues was significantly higher than that in normal tissues (P = 0.001). Conclusion: The expression of hTERT did not change significantly in lymphocytes whether activated or not, and no significant difference with normal tissues.
