金属硫蛋白(MT)是一种能结合各种重金属的低分子量非酶蛋白质。当机体在感染、休克、重金属中毒、炎症和电离辐射等应激条件下,体内合成MT增加,但其病理生理意义尚未阐明。一般认为MT与金属储存、自由基清除及有害物质解毒等功能有关。由于MT对细胞的非特异性保护作用表现在正常组织为减毒,而表现在肿瘤组织则为耐药性,故引起医学界的高度重视。本文就MT的结构、生理学意义、与肿瘤耐药性和抗肿瘤药物毒性的关系,以及对辐射及肿瘤放疗的影响等问题进行了介绍。
Metallothionein (MT) is a low molecular weight, non-enzymatic protein that binds to a variety of heavy metals. When the body in the infection, shock, heavy metal poisoning, inflammation and ionizing radiation and other stress conditions, the synthesis of MT increased in vivo, but its pathophysiological significance has not been elucidated. Generally believed that MT and metal storage, free radical scavenging and detoxification and other functions related. As MT non-specific protective effect of cells in normal tissue is attenuated, while the performance of tumor tissue is drug-resistant, it attracted the medical community’s attention. This article describes the structure of MT, the significance of physiology, the relationship between the drug resistance of tumor and the toxicity of anti-tumor drug, and the influence of radiation and tumor radiotherapy.
