目的观察胞嘧啶脱氨酶(cytosine deaminase,CD)基因和单纯疱疹病毒胸苷激酶(herpes simplex virus thymidine kinase,HSV-tk)基因对胆管癌细胞和胆管癌移植瘤的抑制作用以及转染缝隙连接蛋白43(connexin43,Cx43)基因对旁观者效应的影响。方法四甲基偶氮唑蓝(MTT)法比较双自杀基因系统作用下和/或Cx43基因转染后QBC939细胞的细胞抑制率及旁观者效应。前体药物应用前后测量移植瘤的大小判断自杀基因系统的作用和Cx43蛋白对旁观者效应的影响。结果CD和tk基因在转染细胞中获稳定表达。mRNA和蛋白水平均显示CD+tk+Cx+细胞的Cx43的表达量增加。自杀基因系统对CD+tk+Cx+细胞组和它生成的移植瘤组的抑制作用均比CD+tk+细胞组高。结论双自杀基因系统在体内外对胆管癌细胞和移植瘤均有明显的抑制作用,Cx43基因的导入可明显增强旁观者效应和对肿瘤的抑制作用。
Objective To observe the inhibitory effect of cytosine deaminase (CD) gene and herpes simplex virus thymidine kinase (HSV-tk) gene on human cholangiocarcinoma and cholangiocarcinoma xenografts, Effect of connexin43 (Cx43) gene on bystander effect. Methods MTT assay was used to compare the cell inhibitory rate and bystander effect of double suicide gene system and / or Cx43 gene transfection in QBC939 cells. The effect of suicide gene system and the effect of Cx43 protein on bystander effect were determined by measuring the size of xenografts before and after the application of prodrugs. Results CD and tk genes were stably expressed in transfected cells. Both mRNA and protein levels showed an increase of Cx43 expression in CD + tk + Cx + cells. Suicide gene system on CD + tk + Cx + cell group and its generation of xenograft tumor inhibition were higher than CD + tk + cell group. Conclusion Double suicide gene system has obvious inhibitory effect on cholangiocarcinoma cells and xenografts in vitro and in vivo. The introduction of Cx43 gene can significantly enhance the bystander effect and inhibit tumor growth.
