目的研究防御素(HNP_(1-3))、白细胞介素-8(IL-8)在溃疡性结肠炎(UC)结肠组织中的表达分布以及与UC病变范围和病变程度的关系,探讨其在UC发病机制中的作用;明确IL-8与HNP_(1-3)的相关性。方法用免疫组化法检测36例活动期UC及30例正常对照石蜡包埋组织中HNP_(1-3)及IL-8的表达情况。结果UC组HNP_(1-3)及IL-8均为阳性表达。对照组均为阴性或弱阳性表达。两者在UC组的表达与对照组相比,差异均有统计学意义(P<0.01)。UC病变范围越广、程度越重,HNP_(1-3)及IL-8表达的阳性率越高。UC患者受累黏膜的HNP_(1-3)和IL-8水平明显高于未受累黏膜。且HNP(1-3)和IL-8表达具有明显的相关性(r_s=0.957,P<0.01)。结论HNP_(1-3)的表达与IL-8呈正相关且均参与了UC的发生和发展;HNP_(1-3)可能在UC局部结肠组织破坏和病理变化中起了更重要的作用;HNP_(1-3)、IL-8反映了疾病的炎症程度,并且可作为病情严重程度评定的指标。
Objective To study the expression and distribution of defensins (HNP_ (1-3)) and interleukin-8 (IL-8) in colonic mucosa of ulcerative colitis (UC) and its relationship with the extent of lesion and pathological changes of UC In the pathogenesis of UC; clear IL-8 and HNP_ (1-3) correlation. Methods Immunohistochemistry was used to detect the expression of HNP_ (1-3) and IL-8 in 36 cases of active stage UC and 30 cases of normal control paraffin-embedded tissues. Results The expression of HNP_ (1-3) and IL-8 in UC group were all positive. The control group were negative or weakly positive expression. The expression of both in UC group and control group, the difference was statistically significant (P <0.01). The larger the extent of UC lesions, the more severe the higher the positive rate of HNP_ (1-3) and IL-8 expression. The levels of HNP_ (1-3) and IL-8 in the mucosa of UC patients were significantly higher than those in the non-affected mucosa. There was a significant correlation between HNP (1-3) and IL-8 expression (r_s = 0.957, P <0.01). Conclusions The expression of HNP_ (1-3) is positively correlated with IL-8, and both of them are involved in the occurrence and development of UC. HNP_ (1-3) may play a more important role in the destruction of UC and histopathological changes. HNP_ (1-3) IL-8 reflects the degree of inflammation of the disease and can be used as an indicator of disease severity.
