目的探讨耐力运动和p53抑制剂Pifithrin-μ对ICR小鼠腓肠肌自噬相关基因转录的影响。方法将24只雄性ICR,28周龄小鼠,体重(45±5)g随机分成对照组(C组),Pifithrin-μ组(P-μ组),Pifithrin-μ耐力训练组(P-μ训练组),每组8只。Pifithrin-μ组和Pifithrin-μ耐力训练组均进行皮下注射Pifithrin-μ制剂,其中Pifithrin-μ耐力训练组进行4周跑台训练。经过饲养,末次运动结束12~24 h内处死各组小鼠,Real-time PCR测定鼠腓肠肌p53 mRNA、Beclin-1 mRNA、LC3 mRNA和Atg7mRNA水平进行比较。结果 (1)P-μ组与C组比较p53mRNA、LC3 mRNA、Atg7 mRNA(P>0.05)的表达均无显著性差异,P-μ组Beclin-1 mRNA的表达极显著高于C组(P<0.01);(2)P-μ训练组p53mRNA表达显著高于C组,P-μ训练组Beclin-1 mRNA、LC3 mRNA、Atg7 mRNA表达均极显著高于C组;(3)P-μ训练组与P-μ组比较p53mRNA无显著性差异(P>0.05),P-μ训练组Beclin-1 mRNA表达极显著高于P-μ组(P<0.01),P-μ训练组Atg7 mRNA、LC3 mRNA表达显著高于P-μ组P<0.05。结论在Pifithrin-μ和耐力训练的共同干扰下,骨骼肌中细胞自噬呈增强趋势,其中叠加了耐力训练干扰因素比单纯注射p53抑制剂因素的分组增强趋势明显;其机制是否通过运动激活p53线粒体自噬分子通路尚需要进一步的深入研究。
Objective To investigate the effects of endurance exercise and p53 inhibitor Pifithrin-μ on autophagy-related gene transcription in gastrocnemius muscle of ICR mice. Methods Twenty-four male ICR and 28-week-old mice were randomly divided into control group (C group), Pifithrin-μ group (P-μ group) and Pifithrin-μ endurance training group Training group), 8 in each group. The Pifithrin-μ group and the Pifithrin-μ endurance training group were subcutaneously injected with Pifithrin-μ preparations, and the Pifithrin-μ endurance training group was subjected to treadmill training for 4 weeks. After feeding, mice of each group were sacrificed within 12-24 h after the end of the last exercise, and the mRNA expression of Beclin-1 mRNA, LC3 mRNA and Atg7 mRNA in the gastrocnemius muscle were measured by Real-time PCR. Results (1) There was no significant difference in the expression of p53 mRNA, LC3 mRNA and Atg7 mRNA between the P-μ group and the C group (P> 0.05). The expression of Beclin-1 mRNA in the P-μ group was significantly higher than that in the C group <0.01). (2) The expression of p53 mRNA in P-μ training group was significantly higher than that in C group. The expressions of Beclin-1 mRNA, LC3 mRNA and Atg7 mRNA in P-μ training group were significantly higher than those in C group The expression of Beclin-1 mRNA in P-μ training group was significantly higher than that in P-μ group (P <0.01), and the expression of Atg7 mRNA in P-μ training group was significantly higher than that in P-μ training group , LC3 mRNA expression was significantly higher than P-μ group P <0.05. Conclusion Pifithrin-μ and endurance training co-interfere with the increase of autophagy in skeletal muscle, which superimposed the endurance training intervention factors than the simple injection of p53 inhibitor factor grouping trend of enhancement; its mechanism whether through the activation of p53 Mitochondrial autophagic molecular pathway still needs further study.
