目的观察亚硒酸钠对人急性髓系白血病HL-60细胞系的诱导分化作用。方法HL-60细胞在含8×10-6mol/L亚硒酸钠的RPMI1640培养液中置37℃、5%CO2、95%空气、饱和湿度培养5天,于第5天查细胞形态学及硝基四氮唑蓝(NBT)试验。结果硒在上述条件下可使HL-60细胞形态学分类中较成熟细胞的比例增高(P<0.05),NBT试验阳性率增高(P<0.01)。结论硒在一定条件下可能有潜在的诱导分化作用
Objective To observe the induction and differentiation of sodium selenite on human acute myeloid leukemia HL-60 cell line. Methods HL-60 cells were cultured in RPMI1640 medium containing 8 × 10-6mol / L sodium selenite at 37 ℃, 5% CO2, 95% air and saturated humidity for 5 days. Cell morphology and Nitroblue tetrazolium blue (NBT) test. Results Selenium could increase the proportion of mature cells in morphology of HL-60 cells under the above conditions (P <0.05), and increase the positive rate of NBT assay (P <0.01). Conclusion Selenium may potentially induce differentiation under certain conditions
