用山莨菪碱(654-2)防治油酸引起的大鼠离体肺水肿性肺损伤,观察654-2对离体肺系数,肺通透指数,肺平均动脉压及肺静脉流出液中MDA,SOD,LDH含量的影响。结果表明:654-2可使离体肺系数及肺通透指数降低(P<0.01),MDA产生及LDH释放减少(P<0.01),SOD活性受到保护(P<0.01)说明654-2减轻肺损伤的作用与抗氧自由基有关。但进一步体外实验表明,各浓度654-2不能直接清除黄嘌呤—黄嘌呤氧化酶产生的氧自由基。上述结果提示,654-2可减轻油酸所致离体肺损伤,其作用可能在于稳定细胞膜。
Anisodamine (654-2) was used to prevent oleic acid-induced pulmonary edema in rats with pulmonary edema. The effects of 654-2 on pulmonary coefficient, pulmonary permeability index, mean pulmonary artery pressure and pulmonary venous effusion were observed. SOD, LDH content. The results showed that: 654-2 could reduce the pulmonary coefficient and pulmonary permeability index (P <0.01), reduce MDA production and LDH release (P <0.01), and protect SOD activity (P <0.01) The role of lung injury and antioxidant free radicals. However, further in vitro experiments showed that each concentration of 654-2 can not directly eliminate the oxygen free radicals produced by xanthine-xanthine oxidase. These results suggest that 654-2 can relieve oleic acid induced lung injury in vitro, the role may be to stabilize the cell membrane.
