建立指甲中氯氮平的液相色谱-串联质谱测定方法,并考察长期用药的精神疾病患者指甲中氯氮平的存在情况。指甲经冷冻研磨处理后,利用乙腈-20 mmol.L1乙酸铵缓冲液(7∶3)超声提取。采用Ultra IBD柱分离,二级质谱多反应监测模式(MRM)检测。指甲中氯氮平在0.5~50 ng.mg1内线性良好(r>0.999 9),日内和日间精密度均小于7%,准确度范围为97.4%~99.6%。16名长期用药的精神疾病患者指甲中氯氮平浓度为1.60~14.14 ng.mg1;其去甲代谢物亦被检出。指甲中氯氮平浓度与毛发存在一定相关性(r2=0.971),并略低于同源个体的毛发中氯氮平浓度。
To establish a method for the determination of clozapine in nails by liquid chromatography-tandem mass spectrometry, and to investigate the presence of clozapine in the nails of patients with long-term mental illness. Nails were cryo-polished and then extracted ultrasonically with acetonitrile-20 mmol. L1 ammonium acetate buffer (7: 3). Ultra IBD column separation, secondary mass spectrometry multiple reaction monitoring mode (MRM) detection. Clozapine in nails had good linearity (r> 0.999 9) within 0.5-50 ng · mg -1 with intra-day and inter-day precision less than 7% and accuracy ranging from 97.4% to 99.6%. 16 long-term use of mental illness nails nail clozapine concentration of 1.60 ~ 14.14 ng.mg1; its normetabolites were also detected. Clozapine concentration in nails was correlated with hair (r2 = 0.971), and slightly lower than clozapine concentration in hair of homologous individuals.
