目的:建立人HNE-1细胞株鼻咽癌的BALB/c裸鼠肿瘤模型,观察水疱口炎病毒(VSV)有无诱导鼻咽癌凋亡作用。方法:收集体外培养的人鼻咽癌HNE-1细胞,将HNE-1细胞接种到小鼠右侧后背部皮下,每只接种6×106个细胞(0.1ml)。建立人HNE-1鼻咽癌BALB/c裸鼠肿瘤模型,予以VSV 1×1011pfu/L(plaque forming unit,pfu,噬斑形成单位)治疗和盐水对照组,Hoechst 33258染色荧光显微镜检测小鼠肿瘤病理切片中肿瘤细胞凋亡情况。结果:人HNE-1鼻咽癌BALB/c裸鼠肿瘤模型予以VSV治疗和不治疗相比较,治疗组小鼠的残存肿瘤中凋亡细胞明显增多。结论:通过人HNE-1鼻咽癌BALB/c裸鼠肿瘤模型的体内实验结果表明,VSV治疗能诱导人鼻咽癌HNE-1肿瘤细胞凋亡。
Objective: To establish a tumor model of BALB / c nude mice with human HNE-1 cell line NPC and observe the effect of vesicular stomatitis virus (VSV) on the apoptosis of nasopharyngeal carcinoma. Methods: HNE-1 cells were collected from human nasopharyngeal carcinoma cells in vitro. HNE-1 cells were inoculated into the right back of the mice and subcutaneously in the back, each inoculated with 6 × 106 cells (0.1 ml). The tumor model of HNE-1 nasopharyngeal carcinoma BALB / c nude mice was established and treated with VSV 1 × 1011pfu / L (pfu, plaque forming unit) and saline control group, and Hoechst 33258 staining fluorescence microscope was used to detect tumor Apoptosis of tumor cells in pathological sections. Results: Compared with the untreated VSV group, the number of apoptotic cells in the surviving tumor of the HNE-1 nasopharyngeal carcinoma BALB / c nude mice models was significantly increased. CONCLUSIONS: In vivo experiments in a human HNE-1 nasopharyngeal BALB / c tumor model in nude mice suggest that VSV treatment induces apoptosis in human nasopharyngeal carcinoma HNE-1 tumor cells.
