目的探讨基底细胞癌(BCC)、鳞状细胞癌(SCC)和日光性角化病(AK)组织中PinX1蛋白和人端粒酶逆转录酶(hTERT)mRNA的表达及其意义。方法选取2010年3月至2014年9月间在西安交通大学第一附属医院就诊的49例皮肤肿瘤患者作为观察组,其中BCC患者16例、SCC患者18例、AK患者15例。另选取15例正常人皮肤组织作为对照组。采用半定量逆转录聚合酶链反应(RT-PCR)法,检测BCC、SCC及AK患者组织和15例正常人皮肤组织中PinX1和hTERT的mRNA表达水平,并检测其中的端粒酶活性。结果 PinX1 mRNA在3种不同皮肤肿瘤组织中的相对表达量较正常对照组低,差异有统计学意义(P<0.05),但在3种不同皮肤肿瘤组织之间的相对表达量比较差异无统计学意义(P>0.05)。hTERT mRNA在3种不同皮肤肿瘤组织中的相对表达量较正常对照组高,差异有统计学意义(P<0.05),但在3种不同皮肤肿瘤组织之间的相对表达量比较差异无统计学意义(P>0.05)。结论 PinX1的下调及hTERT上调可能与皮肤肿瘤形成过程中端粒酶激活性及维持机制有关,二者在基底细胞癌、鳞状细胞癌细胞凋亡方面可能存在拮抗作用,检测PinX1和hTERT的表达水平,对评价皮肤肿瘤预后有重要的临床价值。
Objective To investigate the expression of PinX1 protein and human telomerase reverse transcriptase (hTERT) mRNA in basal cell carcinoma (BCC), squamous cell carcinoma (SCC) and solar keratosis (AK) tissues and their significance. Methods Forty-nine patients with skin cancer treated at the First Affiliated Hospital of Xi’an Jiaotong University from March 2010 to September 2014 were selected as the observation group, including 16 patients with BCC, 18 with SCC and 15 with AK. Another 15 cases of normal human skin tissue as a control group. The mRNA expression of PinX1 and hTERT in BCC, SCC and AK tissues and 15 normal human skin tissues were detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR), and the telomerase activity was detected. Results The relative expression level of PinX1 mRNA in three different skin tumors was lower than that of the normal control group (P <0.05), but there was no statistical difference in the relative expression of PinX1 mRNA between the three different skin tumor tissues Significance (P> 0.05). The relative expression of hTERT mRNA in the three different skin tumors was higher than that of the normal control group (P <0.05), but there was no significant difference in the relative expression of hTERT mRNA among the three different skin tumors Significance (P> 0.05). Conclusions The down-regulation of PinX1 and the up-regulation of hTERT may be related to the telomerase activation and maintenance mechanism in the process of skin neoplasm formation. Both of them may play an antagonistic role in the apoptosis of basal cell carcinoma and squamous cell carcinoma, and detect the expression of PinX1 and hTERT Level, the evaluation of the prognosis of skin tumors have important clinical value.
