为大批量尿样本测定肌酐提供简便、准确的实验方法,并实现尿肌酐检测完全自动化。将全自动生化仪苦味酸法肌酐检测中样本参数设定从24μl减至8μl,不改变原有试剂量,使试样比增大3倍。与未修改样本量的试验方法在检测线性、准确度、稀释尿样的肌酐含量,进行比较。结果显示,试验方法与对比方法检测尿肌酐结果比较,差异无统计学意义;减少样本量后检测线性增宽,有利于尿肌酐的检测。提示,苦味酸法测定尿肌酐在试样比约为30∶1时能将检测线性范围扩大到0~2g/L,且检测线性及准确率均较高(R2=0.999 8,平均回收率为101.5%),超出2g/L可设置样品2倍预稀释模式下自动重测,从而实现尿肌酐无需人工预稀释,检测全自动化。

The results showed that the linearity and accuracy of detection of urinary creatinine with picric acid method at a sample ratio of about 30:1 can be increased to 0-2 g / L with a linearity and high accuracy (R2 = 0.999 8, the average recovery was 101.5%), 2g / L can be set to automatically re-test the sample 2 times the pre-dilution mode, in order to achieve urine creatinine without artificial pre-dilution, the test fully automated.
