目的 动态观察在乳癌发生过程中端粒酶活性的变化 ,明确端粒酶在乳癌发生中的意义。 方法 采用 7,12 二甲基苯丙蒽 (DMBA)诱导雌性Wistar大鼠乳癌 ,TUNEL法检测凋亡细胞 ,RT PCR ELISA法检测端粒酶活性。 结果 在乳癌发生过程中端粒酶活性逐渐增强 ,乳腺增生组端粒酶活性高于正常乳腺组 (P <0 0 1) ,乳癌组端粒酶活性高于乳腺增生组 (P <0 0 1)。单发乳癌组与多发乳癌组间端粒酶活性无明显差异 (P >0 0 5 ) ,乳腺增生组织中凋亡细胞多于乳癌组织中凋亡细胞 (P <0 0 1)。 结论 在乳癌发生过程中 ,乳腺凋亡细胞逐渐减少 ,端粒酶活性逐渐增强
Objective To observe the changes of telomerase activity in the process of breast cancer and clarify the significance of telomerase in breast cancer. Methods Female Wistar rats were induced by 7,12-dimethylbenzoflavin (DMBA). Apoptotic cells were detected by TUNEL method and telomerase activity by RT-PCR. Results The telomerase activity gradually increased in breast cancer, telomerase activity in breast hyperplasia group was higher than that in normal breast group (P <0.01), and telomerase activity in breast cancer group was higher than that in breast hyperplasia group (P <0.01) ). There was no significant difference in telomerase activity between single breast cancer group and multiple breast cancer group (P> 0.05). The number of apoptotic cells in breast hyperplasia tissues was more than that in breast cancer tissues (P <0.01). Conclusions In the process of breast cancer, apoptotic cells in mammary glands decrease and telomerase activity gradually increases
